| 【Objective】: To investigate the therapeutic effect of embolization with Apatinib loaded iodized oil on the treatment of hepatocellular carcinoma(HCC)model.【Materials and Methods】: 1.100 rabbits with VX2 liver cancer model were established as well as confirmed of implanted with tumor by enhanced CT.Then they were randomly divided into four groups,which were respectively named as Group I+A,Group I,Group A,Group NS,and each group included25 rabbits): Group I+A(0.1ml/kg iodized oil containing 2mg/kg apatinib,TACE),Group I(0.1ml/kg ultra liquid iodized oil),Group A(the mixture of 0.1ml/kg normal saline solution with 2mg/kg apatinib),Group NS(0.1ml/kg normal saline solution).2.Ten rabbits of each group were scanned by enhanced CT 7 days later after the operation.And the tumor growth rate were calculated,the pathological examination were performed as well.3.On day1,day3,day7 after TACE procedure,the tumor samples of each group(n=15)were harvested,and the expression of MVD、VEGF、HIF-1a were evaluated by using the immunohistochemical staining.【Results】: The TACE procedure were successfully performed in all experimental rabbits.The observation period of 7 days after embolization,the tumor growth rate of Group I+A was significantly lower than other three groups of rabbits(P=0.000<0.01).Immunohistochemistry results showed that the microvessel density(MVD)of apatinib loaded lipiodol embolization group(Group I+A)was significantly lower than that of the other three groups of rabbits(P<0.01).The expression of HIF-1a and VEGF in Group I+A and Group I are also in a dynamic change.On 3 days after TACE procedure,the expression of HIF-1a and VEGF of Group I+A and Group I were higher than day1 and day7,while the expression of HIF-1a and VEGF in Group NS and Group A showed no significant change.【 Conculsions 】 : Embolization with apatinib loaded lipiodol on the treatment for rabbit VX2 liver tumor is simple and feasible,and can effectively inhibit the tumor vessels after embolization as well as inhibiting tumor recurrence and metastasis,which in turn showed a significant inhibitory effect for tumor growth. |
