At present,the tumor is one of the most dangerous diseases that threaten human life.Therefore,early screening and prevention of cancer was important for life medical testing.Recent studies have shown there are the depositions of fibrin in the tumor stroma,therefore the fibrin was considered as a potential marker of cancer,and the early screening of cancer could be achieved by detecting fibrin.The project developed a new strategy for detection of fibrin by using the mimetic enzyme,and the homing peptide CREKA could be recognized the fibrin.In this study,the novel,low-cost,effective and convenient methods for detection of fibrin were achieved.1.Label-free detection of fibrinogen based on fibrinogen-enhanced peroxidaseactivity of fibrinogen-he min compositeA simple,label-free colorimetric method for the determination of fibrinogen?Fib?in plasma is presented.In the work,it was reported that Fib interacted with hemin to form hemin-Fib composite.Because Fib prevented hemin from the formation of m-oxo-dimers,hemin-Fib composite possesses the excellent peroxidase-like activity.Importantly,the peroxidase-like activity of Fib-hemin was increased with the increasing of Fib.This allows us to utilize the ABTS-H2O2 colorimetric system for quantitative analysis of Fib.The optimized method provided a linear determination range 2.0 pM-100 pM with a correlation of 0.9975,and the regression equation was A=0.18546+0.00066 c?c is the fibrinogen concentration,nM?.The limit of detection for Fib was experimentally determined to be 0.7 p M,based on a signal-to-noise ratio?S/N?of 3.This novel approach provides a rapid,sensitive,cost efficient and robust bioassay for detection of Fib in pathology and clinical applications.2.Reducing background signal of fibrin-he min sensing platform by single-walledcarbon nanotubesIt was reported a novel method for the detection of fibrin based on the construction of fibrin-heme peroxidase.In the presence of carbon nanotubes,the fibrinogen,hemin could be absorbed on the surface of carbon nanotubes,and the fibrin could not be absorbed.Thus,a simple,sensitive and selective method for the detection of fibrin was established.Under the optimal conditions,this method provided a linear determination range from 0.1 nM to 10 nM,and the regression equation was A=0.20433+0.00085 c?c is the fibrin concentration,nM?with a correlation coefficient of 0.9970.The present limit of detection for fibrin was 10 pM.3.Sensitivity detection of fibrin method based on the homing peptideThis work established a novel method for the detection of fibrin,and this method relied upon the recognition of homing peptide with fibrin.Homing peptides were a class of fibrin-specific peptides that have been screened by phage display technology.Horseradish peroxidase?HRP?was labeled on the homing peptide and using the ABTS-H2O2 colorimetric system.This method was high accuracy,specificity and simple.Under the optimal conditions,this method provided a linear determination range from0.05 nM to 5.00 nM,and the regression equation was A=0.3400+0.0330 c?c is fibrin concentration,nM?with a correlation coefficient of 0.9976.The present limit of detection for fibrin was 0.01 nM.4.High sensitive method for the detection of fibrin based on the G-quadruplex-he min DNAzymeIn this work,the homing peptides and the G-quadruplex-hemin DNAzyme were conjugated with nanogold particles?AuNPs?to form a response platform by ABTS-H2O2 colorimetric system.The method could realize the molecular recognition and response signal amplification.Through homing peptides specifically recognized fibrin,a new method for the detection of fibrin was established.Under the optimal conditions,this method provided a linear determination range from 0.01 nM to 8.00 nM,and the regression equation was A=0.0160+0.0990 c?c is fibrin concentration,pM?with a correlation coefficient of 0.9969.The present limit of detection for fibrin was 6pM.
