Objective:Vitamin C(Vc)in the treatment of Immune thrombocytopenia(ITP)patients before and after the peripheral blood of cytotoxic T lymphocyte associated antigen 4(CTLA 4),the expression of T cell subgroup and related cytokines,to observe the Immune status of patients with ITP,further explore the Vitamin C in the Immune mechanism in ITP treatment.Methodology:Collected during October 2018 to January 2015 in lanzhou university,the second hospital of ITP patients onset of example,using the principle of random divided into corticosteroids and vitamin C in treatment group and glucocorticoid treatment group.CTLA-4,Th1 cells,Th2 cells,Th17 cells,and regulatory T cells(Treg)were detected by flow cytometry.Using enzyme-linked immunosorbent assay(ELISA)to detect peripheral blood IFN-?,IL-4,IL-10,IL-17 cytokine levels,using SPSS19.0 software analysis between the two groups of patients before and after treatment group and the expression differences between cells and cytokines,analysis of vitamin C and various clinical indicators,the relationship between T cells and cytokines.Results:1.Flow cytometry was used to detect the expression of CTLA-4 and T cell subsets in patients with newly diagnosed immune thrombocytopenia.,the expression of CTLA-4,Th2,and Treg cells was increased in the experimental group and the control group before treatment,and the expression of Th1 and Th17 cells was decreased..Compared with the control group after treatment,the expression of CTLA-4,Th2 and Treg cells increased in the experimental group,and the expression of Th1 and Th17 cells decreased.Detection of peripheral blood related cytokine expression by ELISA,the expression of IFN-? and IL-17 was decreased and the expression of IL-4 and IL-10 was increased in the experimental group and the control group after treatment,compared the experimental group with the control group,the expression of IFN-? decreased,and the expression of IL-4 and IL-10 increased.2.Statistical analysis was used to analyze CTLA-4 negatively correlated with IFN-? and IL-17 before treatment,and positively correlated with IL-4 and IL-10.Conclusion:1.High-dose vitamin C combined with glucocorticoids is superior to glucocorticoids in the treatment of newly diagnosed immune thrombocytopenia;2.CTLA-4,T cell subsets and related cytokines in the experimental group show significant changes,suggesting that high-dose vitamin C effective mechanism of ITP may be to regulate the cellular immune function;3.The exact mechanism of action of high-dose vitamin C needs further study.
