Objective To explore the expression,localization and distribution changes of protein EHD1 and NGF high affinity receptor(Trk A)in rat spinal cord injury model.To investigate the interaction and regulation relationship between EHD1 and Trk A in both vivo and vitro.To elaborate the influence of EHD1 to neurite outgrowth and discuss the important effect on the recovery of neural function after spinal cord injury.Methods We established rat acute spinal cord injury model using the NYU impactor.The injury segment was T9-10 spinal cord.The evaluation of motor function was performed through the BBB score.The expression of protein EHD1 and NGF high affinity receptor(Trk A)in rat spinal cord were detected by Western blot.The localization and co-localization of EHD1 and Trk A,and their co-localization with neurite markers(MAP2)were detected by immunohistochemistry and immunofluorescence.The interaction between EHD1 and Trk A was detected by Co-immunoprecipitation.We established a neuronal apoptosis model by using glutamate to stimulate primary cultured neurons to imitate the circumstance of SCI,and transfected over expression vector and specific si RNA to detect the relationship between EHD1 and Trk A.Results At day 1 after spinal cord injury,the protein expression of EHD1 and Trk A were the highest and their expression pattern showed as the same.The co-localization of both EHD1 and Trk A with neurons and neurite markers(MAP2)increased,meanwhile,the co-localization of EHD1 with Trk A also increased,which were statistically significant(p<0.05).Conclusions After spinal cord injury,the expression of EHD1 was upregulated and may be involved in neurite outgrowth process,which may be involved in the process of neurite outgrowth after injury and important to the recovery of neural function after injury,providing a new approach to the clinical treatment of acute spinal cord injury.
