| Objective To investigate the protective effect of salidroside on human umbilical vein endothelial cells(HUVECs)injured by high-glucose.Methods HUVECs were cultred in vitro.MTS was used to observe the viability of human umbilical veinendothelial cell in different glucose concentrations;According to the result,The glucose concentration groups of 5.5mmol/L were chosen to be the normal groups,and the glucose concentration groups of 45mmol/L were selected to be the model groups;MTS was used to observe the viability of human umbilical veinendothelial cell in different salidroside concentrations;Then different concentrations of Salidroside were given to HUVECs injured by high-glucose.These cells were divided into the normal groups,the model groups and diferent salidroside concentrations groups(Salidroside low dose groups,Salidroside medium dose groups,Salidroside high dose groups and Salidroside more high dose groups).By measuring the viability of human umbilical veinendothelial cell,the contents of malondialdehyde(MDA)and the activity of superoxide dismutase enzyme(SOD)in cell,the protective effect of Salidroside on injured HUVEC was assessed.Results There was no significant difference in normal human umbilical vein endothelial cells cultured with different concentrations of salidroside(24 h F=0.220,p=0.93;48 h F=0.106,p=0.98).Compared with the model groups,the viability of human umbilical veinendothelial cell was increasing significantly(P<0.05),The contents of intracellular MDA was significantly decreasing(P<0.05),and the activity of intracellular SOD was significantly enhancing(P<0.05)in Salidroside medium dose groups,Salidroside high dose groups and Salidroside more high dose groups;the viability of human umbilical veinendothelial cell was increasing(P<0.05),and the content of intracellular MDA was decreasing in Salidroside low group(P<0.05).Conclusion 1.Glucose concentration of 45mmol/L can be used as the best high glucose concentration where HUVEC was injured by high glucose.High glucose can reduce the cell activity of HUVEC.The damage is achieved by inhibiting the SOD activity of HUVEC and increasing the MDA content of HUVEC.2.Salidroside can restore the viability of HUVEC cells damaged by high glucose and provide protection for cells.The protective effect is achieved by enhancing the SOD activity and reducing the MDA content of HUVEC injured by high-glucose. |
