Objective: To observe the changes of autophagy and the effects for gastric mucosa by detecting the expressions of autophagy-related proteins Beclin-1?LC3-?in the rats' gastric tissues under acute hypoxia.Methods: Sixty male Wister rats aging between 6-8 weeks were randomly divided into experimental group(n =50,Decompression chamber,5000m)and control group(n=10,Xining,2260m).The experimental group randomly divided into 1 d,7 d,14 d,21 d and 28 d five groups according to the different time points.The general morphological changes of gastric mucosal tissues and apoptosis of tissue cells were observed by hematoxylin-eosin staining,the expressions of autophagy-related proteins Beclin-1?LC3-?were detected by immunohistochemistry,the expressions of autophagy-related proteins Beclin-1?LC3-? m RNA were detected by Real-time PCR,the results were analyzed using statistics.Results:(1)The gastric mucosal tissue HE staining showed that the injury levels of gastric mucosal tissue were gradually increased with rats into the simulated plateau environment prolonged,the main pathological features of gastric mucosal gland were gradually destructed,and great amount inflammatory cells were infiltrated,even bleeding and apoptosis were investigated by HE staining.(2)The immunohistochemistry showed that expressions of Beclin-1 and LC3-?protein in experimental group were significantly higher compared with the control group(P<0.05),and with prolonged hypoxia time,expressions of Beclin-1 and LC3-?increased gradually(P<0.05).(3)The Real-time PCR showed that the Beclin-1?LC3-? m RNA expressions of gastric mucosal tissue has the same tendency of protein levels(P<0.05).Conclusions:(1)Plateau hypoxia can caused gastric mucosal injury in rats,and the levels were increased gradually with prolonged hypoxia time.(2)Plateau hypoxia can induced autophagy in gastric mucosal tissue cells.(3)Plateau hypoxia may caused gastric mucosal tissue cells injury in rats by increasing autophagy levels.
