| Parkinson’s disease(PD)is a neurodegeneration disorder closely associated with dopaminergic neuron loss.Many studies suggest that it not only be related to excitotoxin,oxidative stress,autoimmune,apoptosis and other mechanisms are closely related.A new method was extracted from the traditional Chinese medicine Achyranthes bidentata by a unique method,named Achyranthes bidentata(ABPP),by the key laboratory of Jiangsu Province,Nantong University.It is well documented that Achyranthes bidentata polypeptides(ABPP)are potent neuroprotective agents in several kinds of neurons.In the present study,therefore,we proposed that ABPPk might be a potential agents for treating PD by preventing dopaminergic neuron loss.To investigate the effect of ABPPk on the function of DA neurons,the viability of cells was measured by MTT assay,Tunel staining and LDH release in vitro.In addition,we observed the damage of mitochondria during the apoptotic process in cell lines SH-SY5 Y and primary DA neurons by mitochondrial membrane potential staining.In vivo experiment,by MPTP 20mg/kg intraperitoneal injection of C57 BL mice to prepare mouse PD model.Protein expression levels were measured by Western blot and immunohistochemistry and immnostaining.The mRNA levels of inflammatory cytokines were examined by real-time quantitative PCR.Mouse behavioral performances were evaluated by rotarod test and pole test.We find that the application of ABPPk,a purified fraction of ABPP,markedly enhances the viability of both SH-SY5 Y cells and primary dopaminergic neurons treated with several neurotoxic agents.In an in vivo MPTP-induced PD mouse model,ABPPk significantly improves behavioral performances and attenuates tyrosine hydroxylase loss in the SNpc.Furthermore,we show that MPTP-induced astrocyte and microglia activation were largely attenuated by ABPPk,leading to low levels of neuroinflammation.Our data show that ABPPk protects dopaminergic neurons from apoptosis,suggesting that ABPPk could be an effective intervention for treating neuron loss associated disorders such as PD. |
