Efficacy And Adverse Reactions Of Ibrutinib In The Treatment Of Chronic Lymphocytic Leukemia:a Meta Analysis

Objective: To make a meta analysis about efficacy and adverse reactions of ibrutinib on chronic lymphocytic leukemia.Method: These following electronic databases were searched: Pub Med,Embase,Cochrane Library,China national knowledge internet,Wanfang database and Weipu database from the building time to December 1st,2017,in order to gain all the single arm clinical trials about the treatment of ibrutinib monotherapy and the treatment of ibrutinib combined with immunochemotherapeutic drugs(anti-CD20 monoclonal antibody,FCR scheme and BR scheme)on chronic lymphocytic leukemia.The standard of literature evaluation came from The system evaluation method of non-random study One of Chinese journal of evidence-based medicine.The meta analysis is aimed to observed indicators such as the rate of overall remission,the rate of complete remission and the rate of severe adverse reactions(neutropenia,anemia,thrombopenia and infection which are equal to or above grade 3).Results: The overall remission rate of the treatment of ibrutinib combined with immunochemotherapeutic drugs is superior to that of the treatment of ibrutinib monotherapy(93% vs 60%,RR=1.42,95%CI: 1.30-1.55),although the rate of complete remission and the rate of severe adverse reactions are similar.Conclusion: The treatment of ibrutinib combination therapy can improve overall remission rate on chronic lymphocytic leukemia compared with that of ibrutinib monotherapy,but no obvious differences in the rate of complete remission and the rate of severe adverse reactions have been observed.Although,the conclusion needs to be further proved by randomized controlled trials in the future.
