The Therapy Effect Of Microwave Ablation To Hepatic Alveolar Echinococcosis In Rat Model

Objective:To investigate the therapeutic effect of microwave ablation(MWA)to hepatic alveolar echinococcosis(HAE)in rat model.Methods:To establish hepatic alveolar echinococcosis models in 70 rats by laparotomy and injected AE suspension.A total of 24 rats were successfully established after model established 6 months,and randomly divided into control group(n=9)and test group(n=15).The test groups were intervened by MWA,while the control groups were performed laparotomy and suture.The lesion diameters were compared between pre-operation and 2 months after operation.All the rats were sacrificed at 2 months after experimental intervention.Serum IL-2 and IL-10 were detected by enzyme linked immunosorbent assay(ELISA).Pathological changes were observed by hematoxylin-eosin staining(HE).The lesion marginal zone levels of IL-2 and IL-10 were detected by immunohistochemistry staining.Results:1.The successful rate of HAE model was 34.3%(24/70)at 6 months.The study group survived 9 rats(9/15)and the controls were 5 rats(5/9)after MWA therapy 2 months.2.The lesion diameters in the test group pre-operation were 0.84± 0.19 cm and 0.65± 0.13 cm after post-operation 2 months(t=3.12,P<0.05),and in the control group were 0.81± 0.13 cm at 6 months and 1.03 ± 0.18 cm at 8 months(t=-4.75,P<0.05).3.The lesion marginal zone levels of IL-2 in the model test group were significantly higher than in the control group,which levels were respectively 129.88±28.25 and 83.56±15.28(t=5.87,P<0.05).The lesion marginal zone levels of IL-10 in the microwave ablation group decreased in comparison to those in the controls,which levels were respectively 125.39±9.60ng/L and 162.83±16.31(t=-9.38,P<0.05).4.The level of serum IL-2 in the test group and control group were respectively 7914.0±2567.8 ng/L and 5882.7±1781.9ng/L,difference between the two groups had statistical significance(t=3.40,P<0.05).Serum levels of IL-10 in the test group were 95.4±17.4ng/L and 113.8±26.9ng/L in the control group(t’=-3.05,P<0.05).Conclusion:1.In vivo experiments suggest that the electromagnetic microwave produce heat could inhibit the further growth of the HAE lesion.2.The IL-2 levels and IL-10 of the lesion marginal zone and serum after MWA therapy suggest that MWA may inhibit focal growth through immunomodulation.
