Objective: To disscuss the role of autophagy in gastric mucosa tissue injury by plateau hypoxia,and explore the correlation between cell aging and cell autophagy under plateau hypoxia environment by observing the changes of gastric mucosa tissue autophagy and detecting the expressions of cell aging related protease TERT and autophagy specificity protein LC3-?,in order to intervente acute gastric mucosal lesions under plateau hypoxia,and provide new targets for improving cell senescence.Methods:(1)Sixty male Wistar rats were randomly divided into control group(n = 10,xining,2260m)and the experimental group(n = 50,decompression chamber,5000 m),the experimental group were divided into hypoxia 1 d,7 d,14 d,21 d and 28 d five groups according to the different hypoxia exposure time.(2)The general morphological and apoptosis changes of gastric mucosa tissue cells were observed by HE staining;(3)The autophagy changes of gastric mucosa tissue cells were observed by electron microscopy.(4)The expressions of TERT and LC3-?of gastric mucosal tissues were detected by using immunohistochemical staining method.(5)The m RNA expressions of TERT and LC3-?of rats gastric mucosa tissue were detected by Real-time PCR;(6)The TERT and LC3-protein? s and their m RNA expressions of rat gastric mucosa tissue in each groups were compared,and the correlation between TERT and LC3-?were analyzed.Results:tory cell infiltration in rat gastric mucosal tissue,gastric bleeding,and glands structural failure,even the nucleus pycnosis apoptosis,and the apoptosis injur y of gastric mucosal tissue were increased gradually with duration of hypoxia exposing.(2)The electron microscope found that hypoxia caused gastric mucos al epithelial cell swelling,necrosis,cavity surface microvilli disappearing,cell nucleus chromatin agglutination,cytoplasm mitochondria swelling,rough endop lasmic reticulum expansion,and cell autophagy structure can be seen,and autophagy structure were increased gradully with the duration of hypoxia exposi ng.(3)Compared with control group,TERT protein expressions were increased i n the experimental groups(P<0.05),and the expressions were increased grad ully with the prolonging of hypoxia exposuring times.Compared with control group,LC3-?protein expressions were also increased in those of the experi mental groups(P<0.05),and the expressions were increased gradully with the prolonging of hypoxia exposuring times.(4)The changes of TERTand LC3-? m RNA expressions induced by hypoxia were the same trend as that of their protein levels.(5)TERT and LC3-II proteins expression levels were positively correlated(r=0.966 p<0.05);TERT and LC3-II m RNA expression levels were positively correlated(r=0.978 p<0.05).Conclusion:(1)Plateau hypoxia can cause rat gastric mucosal cell apoptosis,the apoptosis injury of gastric mucosal tissue were increased gradually with duration of hypoxia exposing.(2)Plateau hypoxia can induce cell autophagy,and the cell apoptosis of plateau low oxygen may be caused by inducing autophagy,then cause the plateau gastrointestinal diseasess;(3)TERT and LC3-II expressions were positively correlated,we speculate that there are correlation between cell aging and cell autophagy,and TERT may be a key protein between those two physiological process.
