Accurate identification of binding media is significant to the research of ancient panting manufacture,art history and the conservation The ingredient and structure of the buried painting binding media under the influence of multi-factor may has been changed,which bring difficulty to the identification.In this dissertation,GC-MS is applied to analysis the amino acid difference between modern animal glue and the ancient animal glue,and the difference between modern binding media and the microorganism aged binding media.Furthermore,explore the relationship between the ancient binding media and the microorganism aged binding media.(1)Setting the amino working curve by 1-10?L the prepared amino solution.The result show that within the limits,11 kinds of amino acid show fine linear,the average correlation coefficient of the amino acid is 0.995.Using the origin regression straight line for this sample that lower than the limits rang,the average correlation coefficient is 0.991.On the basis of building the working curve,determine the amino acid of 20 blank samples.The detection limit is 0.2335?g and the quantification limit is 0.7784?g,those two limits both at the nanogram levels.It is indicate that the painting could be analyzed by slight damag.(2)Study the amino acid difference between the modern bone collagen and the ancient bone collagen.Employ the traditional manufacture to collect the modern binding media(pig,cow,goat)and the ancient binding media(pig,cow,goat).Analyze the 11 amino acids of the binding media by the setting procedure.What's more,compare the same amino acid in same spice but different binding media by descriptive statistics.And study the location variation by the Principal Components analysis(PCA)in the scattered point figure factors and the reason.The result show that the Ser,Asp and Hyp all decrease significantly,while Pro Glu both increase markedly in buried bone collagen,this change may resulting both from the amino acid itself and the microorganism.(3)In order to explore the reason why amino acids of the buried binding media has been changed,the experiment probe into the influence of the microorganism aging in binding media.Comparing the statistical description of the 11 amino acid difference between modern binding media and the microorganism aged binding media(albumen,yolk,whole egg,milk,animal glue),the result show that Ser is decrease in this 5 binding medias,as well as the Hyp in the animal binding media.While the Pro and the Glu both increase in the 5 binding media.Compare to the experimental result of Part Two,the main change of the amino acid is the same,which explain the reason to some extent.(4)Adopting the modern binding media,the ancient binding media and the microorganism aged binding media as the database,11 cultural relic painting from different place have been analyzed by the GC-MS through the setting analysis procedure,The results show that the binding media of the Terra-Cotta Warriors and Disitianwang from Qianfoya are the animal glue-egg mixture binding media.The binding media of the Qing Dynasty painting,the carriages and horses painting from Shangzhou Site and one of Miaozhuang Enpress from Guangyuan Qianfoya are animal glue.The binding media of another painting from Miaozhuang Enpress and the Wangxi are egg.The Linnan Style paintings are milk.The study also found that the ancient and the microorganism aged binding media are more appropriate for the identification of cultural relic painting through the location on the PCA scatter diagram.
