Objective:By observing the expression pattern of regeneration factor,inflammatory cytokines and chemokines inmouse skeletal muscle contusion repair process,investigate the expression rule of muscle regeneration factor,inflammatory cytokines and chemokines in skeletal muscle contusion repair process and may play a role.Methods:Forty C57 male mice were divided into control group(C,n=8)and muscle contusion group(S,n=32).Subsequently,their gastrocnemius muscles were harvested at Od,Id,3d,7d,14d after injury.Hematoxylinand eosin(HE)stain were used to assess the changes of muscle morphology.In addition,the gene expressionof regeneration regulatory factors and angiogenesis factors was analyzed by real-time polymerase chain reaction.Results:1?Morphology of skeletal muscles showed signs of regeneration at 3d post injury.The maximum amount of regeneration muscle fibers appeared during one week post contusion.Two weeks post-injury morphology of myofibers nearly recovered to normal.2?MyoD and Myogenin were the specific markers of proliferation and differentiation of muscle satellite cells,which were significantly expressed after skeletal muscle contusion(P<0.01).3?The M1 type macrophage marker CD68 mRNA significantly increased in skeletal muscle injury in first days and third days(p<0.01),M2 CD163 and CD206 mRNA macrophage markers were significantly increased in the first days of the injury of skeletal muscle(p<0.01),CD163 mRNA and CD206 mRNA decreased rapidly in skeletal muscle damage after third days continue to increase(p<0.01).4.After injury,compared with group C,the regulatory factors for muscle regeneration(IGF-1,MGF,HGF,TGF-? 1)mRNA increased significantly(P<0.05 or P<0.01),the Myostain mRNA reduced significantly(P<0.01).5.The expression of proinflammatory cytokines(IL-1,IL-6,IL-10)mRNA in skeletal muscle after injury(p<0.01 or P<0.05)6.The expression of chemokines(CCL2,CCL3,CCL8,CXCL9,CXCLIO,CXCL12)mRNA in skeletal muscle after blunt trauma(P<0.01)(CCL5).Conclusion:1?M1 and M2 macrophages play an important role in the repair of skeletal muscle injury induced by inflammatory factors(IL-1,IL-6,IL-10)in skeletal muscle after contusion of skeletal muscle.2?Chemokines CCL2,CXCL10 participate in the chemotaxis of immune cells during the repair of skeletal muscle contusion.3?IGF-1,MGF,HGF,TGF-? 1,MSTN play an important role in the repair of skeletal muscle contusion.
