Study Of Dopamine Change In The Rat Striatum When Voluntary Wheel Running Improves Recovery Of Cognitive Function After Global Cerebral Ischemia

Objective:Physical exercise has been demonstrated to be neuroprotective in ischemia models.However,the exact neuroprotective mechanism needs to be further studied.The purpose of this research is to discuss the effect of pre-ischemic voluntary wheel running on levels of dopamine in the striatum and cognition in ischemia rats.Methods:24 Sprague-Dawley rats were divided randomly in following four groups: 4 weeks running-ischemia group(4RI group,n=6),1 weeks running-ischemia group(1RI group,n=6),ischemia group(I group,n=6)and shame operation group(C group,n=6).After training,global cerebral ischemia was induced by two-vessel occlusion(2-VO).In vivo microdialysis and high performance liquid chromatography system(HPLC)were used to collect and determine dopamine in the dialysates of striatum.Additional 24Sprague-Dawley rats were elected to perform the passive avoidance to test cognitive function 24 h after ischemia,and those rats had same groups and exercise training as described above.Results:1.Comparing with constant striatal dopamine level of S group,the striatal dopamine level of I group was rapid increasing and reached maximum value at the time point of 20 min after ischemia(P<0.001),then decreased gradually.The striatal dopamine level of 4RI and1 RI group were also increased after ischemia and reached maximum value at the time point of 20 min after ischemia,then decreased gradually,and the striatal dopamine level of 4RI and 1RI group were no significant difference between baseline level before ischemia and those after ischemia(P>0.05).2.Compared with S and I group,the base levels of striatal dopamine in 4RI and 1RI group were significant increased(P<0.001).However,the base levels of striatal dopamine beween 4RI and 1RI group didn't have significant difference(P>0.05).3.Behavioral data indicated that the second training time of 4RI and 1RI group were significant longer than I group(P<0.001).However,the second training time of 4RI and1 RI group didn't have significant difference when compared with S group(P>0.05).4.Morphologcial results showed that the microdialysi probes were accurately placed in the stiatum,this indicated that dialysate collected were the extracellular fluid of striatum.Conclusion:4 weeks and 1 week pre-ischemia voluntary wheel running can significantly increase the base levels of striatal dopamine,attenuate the increase of striatal dopamine induced by cerebral ischemia and improve cognitive function in ischemia rats.There may be the mechanism that exercise protect the cognition function of ischemia rats.
