Diet analysis is an analysis on animal feeding species,frequency and manner,traditionally,it relys mainly on the microscopic observation and analysis of feces or intestinal contents.With the establishment and improvement of DNA barcoding database,DNA barcoding technology has been widely used for dietary analysis,which has greatly promoted the identification of dietary species.In this study,93 specimens of grasshoppers from2 sites were collected,which,respectively,were Qinling NanWuTai and Shaanxi Normal University,including Trilophidia annalata,Atractomorpha sinensis,Acrida cinerea,Chorthippus chinensis,Euchorthippus unicolor,Pedopodisma tsinlingensis,Fruhstorferiola omei,Xenocatantops brachycerus and Calliptamus abbreviatus.Partial regions of rbcLa were introduced as DNA barcode to analyze the dietary of 9 grasshopper species,and the detailed feeding species information was obtained.The main results are as follows:(1)The primary feeding family of plants of the 9 grasshopper species is Gramineae,followed by Compositae and Umbelliferae,Rosaceae,Carnation,Liliaceae,Carpaceae,Polygonaceae and other 12 families of plants are also feeded by some grasshoppers.The possible dietary of Trilophidia annalata are families Gramineae,Compositae,Rosaceae and Scrophulariaceae.The possible dietary of Atractomorpha sinensis are families Compositae,Umbelliferae,Scrophulariaceae and other 6 families of plants.The Atractomorpha sinensis mainly feed the Digit aria and other 5 genus of the Gramineae,and 2 genus of Umbelliferae,including Daucus and Anthriscus.The possible feeding of Chorthippuschinensis are 7 genus of Gramineae.The mainly feeding of Pedopodisma tsinlingensis are 3 genus of Caryophyllaceae,including Stellaria,Cerastium and Myosoton.The mainly feeding of Euchorthippus unicolor are 9 genus of the only one family Gramineae.The mainly feeding of Fruhstorferiola omei are 4 families including Liliaceae,Geraniaceae,Umbelliferae and Caryophyllaceae.The mainly feeding of Xenocatantops brachycerus included the Artemisia of Compositae and the Quercus of Fagaceae.The mainly feeding of Calliptamus abbreviates is Compositae,and including other 7 families.(2)The plants fed by locusts are also related to the plant groups grown in their habitats,and locusts dietary are differentiated in different habitats.Atractomorpha sinensis collected on campus not only take the Artemisia of Compositae,but also take Rubiaceae and Scrophulariaceae.But Umbelliferae is excluded,it may be associated with the campus artificial cultivated plants species and habitat on plant distribution.(3)Based on the analysis of the feeding habits of grasshoppers in different periods,the feeding habits of grasshoppers were analyzed on time scale.The analysis of possible feeding results in the early and middle term of Calliptamus abbreviatus include Moraceae and Compositae,and Compositae appeared both in the first,middle and later periods.Through the research above,the plant families and species of 9 grasshoppers species were preliminarily figured out.The results show that barcoding technology has high efficiency and simplicity.The rbcLa barcode can be used as a powerful tool for the analysis of the locusts feeding habits.
