Cellar mud is the basis in strong-flavor liquor production process.The categories and proportion of microbial populations in cellar mud have great impact on the flaour of white liquor.facultative bacteria is the main preponderant flora in cellar mud,so classificate and identificate them is necessary,then ascertain the categories and proportion,and establish the relation between their speciality and the flavour of white liquor.The main reason of high yield,low quality,high sale,low profit of Heilongjiang province white liquor lies in white liquor enterprises lack high qulity liquor cellar.Therefore,it is very important that research the structure,distribution and metabolites of microbial populations in the cellars of Heilongjiang white liquor industry,separate and select functional strains for improving the quality of Heilongjiang province white liquor and establish microbial database of liquor cellars.In this research,cellar mud samples that from different position of two different age cellars of fuyu old cellar spirit Co.,Ltd Heilongjiang Province were gather.The pure culture was obtained from the sample by classical microorganism separation and purification technology,the pure culture was identify and microbial populations of cellar mud in different cellar age and different position were statistical comparative analysised by morphology,physiological and biochemical indexes;select by physiological and biochemical indexes and gas chromatographic analysis(GC),and then the functional strains were indentified to genus by 16S rDNA molecule appraisal.Microbial populations structure of the cellar mud samples were analysised by combining traditional cultivation methods and PCR-DGGE molecule technology,the diversity of microbial populations in the cellar mud were more comprehensive understanding.It was known that bacteria types and quantity in Cellars of different age or in different part of one Cellar have significant differences.Bacillus,Halobacillus,Paenibacillus,Aneurinibacillus,Brevibacillus,pseudomonas,Sporosarcina were known by analysising microbial populations with morphology and Physiology and biochemistry identification in cellar mud,and Bacillus is dominant species amomg them.10 functional strains which high-yielding acid ester were select by physical and chemical analysis and gas chromatographic analysis.Bacillus subtilis,Bacillus licheniformis,Bacillus endophyticus,Bacillus alcalophilus,Pseudomonas aeruginosa,Aneurinibacillus migulanus,Bacillus fusiformi,Bacillus megaterium were determined by 16S rDNA molecule appraisa.The diversity of microbial populations in the cellar mud were analyzed by DGGE(denaturing gradient gel electrophoresis)and 15 different bands were detected,and the abundance of microbe in 10 samples of different parts is different.Microbe in the bottom of Cellar I is the most,7 bands were detected.And 5 bands were detected in the bottom of Cellar ?.The similarity of 10 samples of different parts is different.The most similar of microbial community between the first layer and the third layer in the wall of Cellar ? that similarity reach to 75.7%.The least similarity of microbial community between the second layer of cellar II and the fourth layer in the wall of Cellar I that similarity reach to 6.4%.
