Fluorescence spectroscopy have been widely used in the fields of medical diagnostics, biological engineering and substances analysis. But they are limited by weak signal and poor stability in many practical applications. With the development of synthetic and micro-nano fabrication technology, the researchers found that the fluorescence signal or Raman scattering signal will be significantly enhanced by locating molecules on near metallic nanoparticles or metal surface, and thus improve the detection sensitivity. Therefore, surface enhanced fluorescence and surface enhanced Raman scattering were born at the moment, the study of surface enhanced have been focus on the preparation of new efficiency substrates and the research of enhancement mechanism.In this paper, the preparation of silver nanowires (Ag NWs) were deposited onto silicon substrate generate different loading densities can be used enhancement substrates. The metal enhanced fluorescence effect of FS was investigated with different Ag NWs loading density as well as the different excitation wavelengths. While discussed influence of different loading densities of Ag NWs on surface enhanced Raman scattering(SERS)effect of CV and R6G; further investigation the detection sensitivity of high load amount of Ag NWs and single Ag NWs as substrates.Main content of this paper:1. Ag NWs of high yield and uniform were quick and simple synthesized by the improved conventional polyol method, remain other conditions unchanged, investigated systematically the influence of the molecular weight of PVP on Ag NWs growth, the experimental result shows that the large molecular weight of PVP move conducive to AgNWs growth, and the growth mechanism of Ag NWs was discussed.2. The synthesized of Ag NWs were deposited onto silicon substrate generate different loading densities as surface enhancement substrates. The sandwich sample configuration were used for the fluorescence investigation. Effect of the Ag NWs with different loading densities as well as different excitation wavelength on the fluorescence emission of FS was discussed. The experimental result shows that the MEF effect depends on the loading density of Ag NWs and excitation light obviously.3. Under the excitation wavelength of 532nm as Raman spectroscopy analysis, the influence of different loading densities of Ag NWs on surface enhanced Raman effect of CV and R6G was investigated experimentally, the result show that the high load amount of Ag NWs used as substrate to obtain stronger SERS signals. In the same conditions, CV and R6G is selected as probe molecule, the detection sensitivity of high load amount of Ag NWs as well as single Ag NWs as substrates was investigated experimentally.
