With the development of fluorescence microscopy,fluorescence imaging has become a powerful tool for the detection of complex life system.Small molecule fluorescent dyes are very useful tools for fluorescence imaging,and the effective construction of fluorescent dyes can promote the progress of cell biology and drug diagnostic imaging.The defects of traditional fluorescent dye molecules,such as light resistance and toxicity limits their application in biological imaging in the field.Therefore,it is very important to modify the structure of the classical fluorescent dyes for improving or modifying their defects.In this paper,we designed and synthesised 19 kinds of fluorescent dyes and two fluorescent probes,which are modified by the structure of anthracene ring fluorescent dyes,The performance and its application are studied and mainly includes the following two parts.1.On the synthesis of xanthene ring structure as the main body of the Ral-9,Rb1-8,Rcl-2 and other 19 kinds of novel fluorescent dyes,IR,1H NMR,13C NMR,HRMS(ESI)of the new compounds were characterized;spectroscopic properties show that the dye has excellent photostability,fluorescence emission wavelength red shifted to more than 600nm;It can effectively reduce the defects of autofluorescence interference in fluorescence imaging,and the Stokes shift of the fluorescent dye is 100-120nm,which is higher than that of the traditional anthracene ring fluorescent dyes;DFT,TDDTF and simulated by theoretical calculation,studies the characteristics of fluorescent dye molecular structure,explains why the fluorescence quantum yield increased and the emission wavelength redshift phenomenon;dyes used for Pc 12 staining,cell toxicity,white rat fluorescence imaging showed that Ra,Rb,Rc series of dyes on the cell toxicity is small,can be used for cell imaging,Rb and Rc series of fluorescent dyes on mice showed fluorescence imaging can be used for in vivo imaging.2.The xanthene ring fluorescent dye matrix,can further design of fluorescent probe T1,T2 recognition Hg2+ synthesis;ion selective experiments show that single fluorescent probe with excellent selectivity for Hgig,The interference of coexisting ions indicates that other metal ions have little effect on the recognition of fluorescent probes;interference ions showed little effect on the recognition of other metal ions on the fluorescence probe;fluorescence titration experiments and calculation shows that the fluorescent probe detection limit identification of Hg2+ sensitive fluorescent probe;optimum pH for 7-8,with the application range of the organism;according to NBO(NBO)calculated fluorescence probe molecules with Hg2+ binding sites and the best possible reaction mechanism;cell imaging experiments showed that the fluorescent probe can be applied to the detection of metal ions in cells.It has potential application value in the field of biological detection.
