Affinity membrane separation is a new biological purification technology that combines membrane separation and affinity chromatography technology to achieve specificity and high purification rate adsorption.With the development of preparation technology of nanofiber technology,nanofiber affinity membrane technology has attracted wide attention.As a natural adsorbent,chitosan exhibits good adsorption performance.In this study,chitosan derivative was used as the affinity membrane material,and the nanofiber affinity membrane was prepared by Solution blowing.This will provide new ideas and methods for the study of protein specific adsorption separation.In this paper,firstly,the arginine/chitosan(CS-Arg)was prepared by grafting arginine onto chitosan and using varying molar ratios of arginine to chitosan.And characterized by FTIR,13C NMR,XRD,TG and EA,the results showed that with the increase of arginine content,the substitution degree first increased and then decreased.Furthermore,the nanofiber affinity membranes(CANFs)were prepared by Solution blowing using the prepared CS-Arg and PLA as the blended solutions.The CANFs were characterized by SEM,Contact angle,Filtration efficiency and Resistance,FTIR and XPS,the results showed that with the increase of the degree of substitution,the diameter of the fiber increased slightly and the average diameter was 100-200nm.The CANFs was hydrophilic and had high filtration efficiency and low filtration resistance.Then,bovine serum albumin as a model,study on adsorption of CANFs.The results showed that the adsorption capacity of CANFs was good and the maximum adsorption capacity was 445.19mg/g.The effects of pH,initial concentration and adsorption time on the adsorption of BSA were discussed and the optimum adsorption conditions were pH=7.14,adsorption time 10h and initial concentration 3.0mg/ml.The adsorption of CANFs for BSA was in accordance with Langmuir model,and it was a monomolecular layer adsorption,and the affinity was in the middle range,which was effective adsorption and easy elution.The adsorption rate was in accordance with the quasi-second-order kinetics model,which did not accord with the molecular diffusion model.The molecular diffusion rate control step was chemisorption.The study on the elution of BSA shows that it has a high regenerative adsorption property and can be reused and has good stability.In this paper,the adsorption properties and adsorption models of the nanofibers were studied,which provided the data and theoretical guidance for the development of the new amino acid ligands and the study of the affinity chromatographic separation process,thus broadening the affinity membrane technology applications.
