In this paper,Sarocladium strictum CA022,Acremonium cucurbitacarum AJ013 and Simplicillium obclavatum YX015 respectively were cultivated by solid-state fermentation method,and were crude extracted by five organic solvents with different polarity(dichloro methane,MSOtroleum ether,ethyl acetate,methanol and n-butanol).Finally,antioxidant activities of the crude extract were determined.The results were as follows:1.Three isolates were cultivated by improved rice solid culture.A fixed solid fermentation conditions(conditions for liquid culture of seed cultivation:the speed of vibrator is 130 rpm,temperature 28?;solid fermentation conditions:pH6.8,dark cultured under 28 ?)were adjusted and determined.2.Four methods(?-carotene bleaching method,DPPH method,pyrogallol autoxidation method and salicylic acid method)were be used to determine the total antioxidant activity,DPPH radical scavenging activity,superoxide anion scavenging activity and hydroxyl radical scavenging activity of the three isolates.The total antioxidant activity of crude extracts of S.strictum CA022 and A.cucurbitacarum AJ013 were lower than the positive control-BHT,but S.obclavatum YX015 methanol crude extract activity(inhibition rate was 33.66%)were equal to the activity of BHT(inhibition rate was 30.14%);DPPH radical scavenging rates of the crude extracts were lower than BHT,and there is a significant difference(P<0.05);The three isolates' crude extracts with the highest superoxide anion scavenging activities were crude extracts of dichloromethane(clearance rate was 60.73%),ethyl acetate(clearance rate was 57.44%)and petroleum ether(clearance rate was 68.20%)respectively.Their scavenging activities were all higher than BHT(clearance rate was 24.51%),and there is a significant difference(P<0.05);For hydroxyl radical scavenging activity,the clearance rate of extract which was crude extract by n-butanol(clearance rate was 67.62%)was higer than it of BHT(clearance rate of 59.56%),and there was significant difference(P<0.05),the clearance rate of a crude dichloromethane extract of A.cucurbitacarum AJ013(clearance rate was 62.46%)was equal to it of BHT,but the rate of crude extracts of S.obclavatum YX015 were lower than that of BHT.In conclusion,dichloromethane,methanol and n-butanol can be selected to extract the three isolates to obtain a crude extract with higher antioxidant activity.3.The three extracts of S.strictum CA022 with higher antioxidant activity were be used for HPLC fingerprint study.Chemical compositions of these extracts were got across preliminarily.
