S-(-)-5-difluoromethoxy-2-[[(3,4-dimethoxy-2-pyridinylmethyl)]sul phinyl]]-1H-benzimidazole is the chemical name for S-pantoprazole,which is the single isomeride of the pantoprazole.Compared to its r acemic modification and(+)-pantoprazole,(-)-pantoprazole had better selectivity,higher bioavialbility,longer half-life,better plasma protein binding rate and lower toxic side effect.This study use maltol(3-hydroxy-2-methyl-4-pyrone)as row mate rial,through methylation,ammonization,chlorination,oxidation,meth oxylation,rearrangementreaction,hydrolyzation to obtain the hydroch loride form of the 2-chloromethyl-3,4-dimethoxypyridine.The other si des use Panadol(acetaminophen)as row material to obtain 5-difluor omethoxy-2-mercapto-lH-benzimidazole by herification,nitration,deox idization,ring-closure reaction steps.Then the pantoprazole was synt hesized from those two compounds by condensation.S-(-)-5-difluoro methoxy-2-[[(3,4-dimethoxy-2-pyridiny lmethyl)]sulphiny 1]]-1 H-benzimi dazole was synthesized from(-)-diethyl D-tartrate as the chiral rea gent and cumene hydroperoxide as the oxidizing agent by asymmetr ic oxidation,then get the S-pantoprazole sodium and S-pantoprazole megnesium.through the condensation reaction between S-(-)-5-difluor omethoxy-2-[[(3,4-dimethoxy-2-pyridiny lmethyl)]sulphinyl]]-1 H-benzim idazole and sodium hydroxide,magnesium chloride.Meanwhile synt hesis technology inculed hydrogen peroxide oxidation,methoxylation,rearrangement reaction,hydroxylation,condensation was analyzed,especially the asymmetric oxidation of the S-(-)-5-difluoromethoxy-2-[[(3,4-dimethoxy-2-pyridinylmethyl)]methyl]thio]-1H-benzimidazole,whi ch is the critical step of the synthesis technology has been went int o painstaking detail,we acquire the optimization process.Products detected by HPLC more than 99.5%purity,isomer con tent below 0.1%;structure combined with the synthesis process,HP LC profiles,liquid-qualitative confirmation.The process of raw m aterial is homebred change,suitable for the industrialized production requirement.
