Mold lives everywhere.It is one of the major spoilage microbes in food.Rhizopus nigricans is a fungus root,also known as prostrate branches Rhizopus,mildew often appears in foods,such as fruits and vegetables.In recent years,plant-derived preservative was popular in the public.Scutellaria showed best inhibitory effect against Rhizopus nigricans from more than 20 kinds of plant materials,and the MIC was 8 mg/mL.Extract process was optimized by univariate analysis and the Box-Benhenken response surface experimental design,based on the response surface test and regression analysis.The optimum extraction parameters to skullcap was that the extraction time was 6.17 h,liquid-solid ratio was 9.87:1(mL:g),extraction temperature was 70 ? the concentration of ethanol was 95%.At the same time,inhibition zone diameter against Rhizopus nigricans was 15.64 mm.Cell ultrastructure of Rhizopus nigricans were observed by scanning electron microscopy and transmission electron microscopy.The results showed that black mold root morphology SEM treatment group showed irregular,varying thickness,cell wall rupture,cytoplasmic loss;TEM-treated cell,intracellular vacuole shape changes,but the walls still visible.Changes in content of the extracellular protein,alkaline phosphatase(AKP),conductivity,K+ and inorganic phosphorus,UV absorption material,succinate dehydrogenase(SDH)were analyzed in order to study inhibitory mechanism of the extract against Rhizopus nigricans.The results showed that alcohol extract of Scutellaria destroyed the structure of the cell wall and membrane of Rhizopus nigricans,which leaded to increased membrane permeability;thereby enable the cell contents leak.BOD5 can be fed the microbial respiration intensity.The inhibitor can be added before and after the cell respiration rate,you can find an inhibitor of cell respiration inhibition.Shows data from the overlap rate,while ethanol extract of Scutellaria malonate mixing overlap with the maximum rate,followed by iodine acetic acid,and finally sodium phosphate.This is inferred alcohol extract of Scutellaria maximum inhibition of the Krebs cycle,followed by glycolysis,and finally the pentose phosphate pathway.
