Now nanocapsule has become a new kind of drug delivery system with extensive potential prospect of application.Some drugs with the remarkable curative effect have poor solubility in humen body filled by water,if these drugs can be coated by water-soluble wall material,they could be transported in body conveniently.Drugs after nanocapsulated can increase the stability and cover up the smell.Nanocapsules are long-term drug delivery system with controlled release and low toxicity.In this paper,nanocapsules loading nimodipine were prepared in O/W microemulsion by the complex coacervation with the fat-soluble drugs nimodipine as drug model,chitosan and gelatin as wall material.Nanocapsules were analyzed as drug carrier in morphology,drug embedding,drugs release and related performance.The nanocapsules had good size dispersion and uniform particle size.The size of blank nanocapsules was between 30~50 nm and drug-loading nanocapsules had an average particle size between 70~120 nm.Encapsulation efficiency over 50%can be obtained,the factors on drug encapsulation and drug encapsulation exhibited inverted U-shape,so the optimal preparation conditions of the experiment can be gotten.Nano drug pharmacokinetic study is the prerequisite to clinical application,so human body conditions were simulated.The results show that drug-loading nanocapsules had the clear effects with controlled release compared with the raw material,prolonging the time from 200 minutes to 700 minutes corresponding to the cumulative release dosage of 90%.Biological compatibility is the key of biological materials entering the clinical research.The cell morphological analysis was determined by MTT experiment to evaluate the cell toxicity of drug-loading nanocapsules.It turned out that toxic effects of drugs on cells were reduced after nanoencapsulation.Uniform size and controllable size distribution of nimodipine nanocapsules were prepared in this paper,with high entrapment efficiency,good release performance,and good biocompatibility.A theoretical basis was provided for the further research and application of drug loaded nanocapsules.
