Rapeseed is one of the most important oil crops in China.It is not only an importa nt source of oil and feed,but also an important industrial raw material.For a long time,the improvement of the main quality traits such as oil content,fatty acid compositions and glucosinolate content of rapeseed has been the main target of rape breeding at home and abroad.Rapeseed seeds have significant secondary dormancy properties,and this ch aracteristic is an important reason for the generation of seedlings(weeds)and gene diffus ion.Therefore,the combination of rape quality breeding and secondary dormancy breedin g has become the current direction of rapeseed breeding.The current development of rap eseed(Brassica napus L.):The results showed that the fatty acid metabolic pathway gen e was significantly enriched after induction,and ABA,GA and other hormone synthesis and signal transduction pathway related gene expression,and ABA,GA and other hormo nes had an important effect on the formation of the main quality of rapeseed,suggesting that there was a certain correlation between secondary dormancy characteristics and mai n quality.However,the correlation between oil content,fatty acid,glucosinolates conte nt and secondary dormancy characteristics of rapeseed has been largely unknown.In this study,Brassica napus cultivars preserved in the laboratory were first used to analyze th e differences between the main quality traits and the secondary dormancy characteristics among the cultivars;second,the correlation between the main qualities and that between quality and secondary dormancy characteristics was clarified by means of Nutrition,Gene tic breeding and Molecular biology;third,we established a high quality and rapid extract ion method for mature seeds of rape to provide technical support for further analysis of the genetic basis of main quality and secondary dormancy characteristics.The main findi ngs are as follows:(1)PEG-induced rapeseed seed dormancy results showed that the dormancy level wa s different under the same conditions,in which the highest rate of dormancy rate was 98%,the lowest was 6%.(2)The analysis of oil content,fatty acid and glucosinolates showed that the other c ultivars were close to 40%except the gra 4.Fatty acid content analysis found that oleic acid,linoleic acid and erucic acid among different rapeseed varieties showed significant differences.Five out of the national stardard for low glucosinolates and the content in gr a 4 was the highest by reaching up to 143.09 μmol/g.(3)The correlation between secondary dormancy oil content,fatty acid and glucosin olates analysis showed that secondary dormancy and the oil content at the 0.05 level th ere is no significant correlation;Only under the condition of high dormancy rate(>80%),secondary dormancy and palmitic acid at the 0.05 level was significantly negative correl ation(r=0.987),and the rest of the fatty acid composition at the 0.05 level there is no s ignificant correlation.(4)The correlation analysis of different components of fatty acids showed that there were different degrees of correlation between different components of fatty acids:myrist ic acid was positively correlated with sevatric acid;palmitic acid negatively correlated wi th peanutoic acid and erucic acid,and positively correlated with stearic acid,oleic acid,l inoleic acid and peanutenoic acid;heptadeca-enoic acid negatively correlated with peanute noic acid,peanutinic acid,Behenic acid,erucic acid and tetracosenoic acid,and positivel y correlated with stearic acid and oleic acid;stearic acid negatively correlated with pean utoic acid,peanutenoic acid,behenic acid,erucic acid,tetradecenoic acid,and positively with oleic acid;there was a significant negative correlation between oleic acid and arachi die acid and tetracosonic acid,and oleic acid highly significantly negatively correlated wi th peanutenoic acid,peanutenoic acid,behenic acid,erucic acid and tetracosenoic acid;I inoleic acid significantly positively correlated with linolenic acid and erucic acid,and neg atively correlated with peanutoic acid and peanutenoic acid;arachidic acid positively corr elated with peanutoic acid,peanutenoic acid,behenic acid,erucic acid and tetracosenoic acid and significantly positively correlated with tetracosenoic acid;peanutenoic acid positi vely correlated with peanutenoic acid,beech acid,erucic acid,tetracosenoic acid,and pea nutenoic acid acid positively correlated with behenic acid,erucic acid,tetracosenoic acid;erucic acid positively correlated with tetracosenoic acidacid。(5)The correlation between oil content and fatty acid showed that the oil content w as positively correlated with hexadecanoic acid and oleic acid,and negatively correlated with the content of arachidic acid,peanutenoic acid,behenic acid,erucic acid,tetracosen oic acid.(6)The results of correlation analysis between glucosinolates and fatty acids showed that glucosinolate significantly negatively correlateded with palmitic acid and linoleic aci d,and positively correlated with the arachidic acid;glucosinolate highly significantly neg atively correlated with the contents of myristic acid,stearic acid,oleic acid and peanutoic acid,and highly significantly positively correlated with peanutoic acid,peanutenedioic ac id,behenic acid,erucic acid and tetracosenoic acid.(7)The combination of RNA plant plus reagent of TIANGEN and RNA Plant Kit(DNase I)of KANGWEI applied to exteact the mature rapeseed rich in polyphenols,fats and proteins.The extraction time can be shortened to prevent RNA degradation,and the concentration and quality of RNA could meet the molecular detection.(8)In terms of nutrition indexes,gra3 is suitable for promoting the cultivation,which has h igher oil content(>60%),higher linoleic acid,lower linolenic acid and glucosinolates content(<1%),ensuring high yield eff-iciency as well as the taste;combined nutrition index and seconday dormancy trait,gra 7 is more suitable because lower seed dormancy trait is beneficial to reduce t he occurrence of volunteers,thus ensuring the stability of the quality of rapeseed.
