Objective To study the expressions of adrenomedullin (ADM) protein and mRNA in human tongue squamous cell carcinoma (HTSCC) and analyze its relationship with angiogenesis, provide new target and experimental basis for anti-angiogenesis treatment of HTSCC.Methods1.The expressions of ADM and VEGF protein were measured by immunohistochemical assay in35HTSCC tissues, and make micro vessel density (MVD) count. Analyzed the expression of ADM protein in HTSCC tissues and its relationship with clinical pathological characteristics, and analyzed the relationships among ADM and VEGF, MVD counts.2. Using real-time fluorescent quantitative RT-PCR assay for detection of ADM and VEGF mRNA express in33cases of HTSCC tissues. Statistical analyzed the expression of ADM mRNA and its relationship with clinical pathological parameters, and analyze the correlation between ADM and VEGF expression.Results1.The expressions of ADM mRNA and protein in HTSCC tissues.ADM protein was primarily localized in the cytoplasm of the carcinoma cells. ADM mRNA and protein expressed in most of the HTSCC tissues, The positive rates of ADM in HTSCC were significantly higher than normal tongue tissues(/=2.203,P<0.05;Z=-5.441, P<0.01).2. The relationship between ADM expression and clinicopathological features in HTSCCThe expression of ADM significantly associated with histological grading (F=37.132, H=4.212;F<0.05), the size of the tumor (t=1.532,Z=-3.398; P<0.05) and lymph node metastasis (t=1.571,Z=-2.604; P<0.05).3. The relationships among ADM and VEGF, MVD countsThere were positive correlations between ADM and VEGF mRNA and protein expressions(r=0.776,0.623; P<0.01).ADM also had a positive relationship with MVD count (r=0.772; P<0.01)Conclusion It has shown for the first time that ADM expressed in HTSCC, and it may induce tumor angiogenesis through promote the expression of VEGF, and then promote the growth and metastasis of the tumor. It may be that ADM as a new molecular target to provide new research and treatment strategies for HTSCC treatment.
