Objective: To observe Rougantongluo Decoction's effect of XIAP/Smac on the expression of MCAO in rats after reperfusion.Methods: 96 male Sprague-Dawley rats were randomly divided into four groups: the control group, the sham-operated group, the MCAO model group, and the Rougantongluo Decoction group.We performing intragastric administration with 1.4ml/100 g saline to groups except of the Rougantongluo Decoction model group. Preparation of distilled water by 14ml/kg perfusion in rats. We cotinuous pretreating the experiment object through drug administration twice a day and given 30 min before MCAO operation for the last time.Samples in all groups except those in the control group were duplicated into models of MCAO with Zea-longa modified thread-occlusion method. After MCAO successfully established, neurological function deficits were observed at each time(3 h/6 h/12 h/24 h/48h)after cerebral ischemia-reperfusion.Morphological changes of brain tissue was measured by TTC staining method on 24 h reperfusion,cell apoptosis was detected by TUNEL method,the expression of XIAP and Smac around the infarct was detected by immunohistochemical staining(SP).Results:1.Neurological defect symptoms of MCAO rats were obviously ameliorated in Rougantongluo Decoction(P<0.05), especially after 3 h/48 h reperfusion. Changes of infarct area were consisted with neurological function deficits score.2.The TUNEL assay showed that the apoptosis cells surrounding the infarcted area of MCAO rats were obviously increased,which has statistical significance campared with the contorl group and the sham group(P<0.05),while the apoptosis cells of Rougantongluo Decoction was significantly decreased compared with the model group(P<0.05).3.Immunohistochemical staining showed that XIAP and Smac were obviously expressed surrounding the infarcted area of MCAO rats. The expression of XIAP was significantly increased while the expression of Smac protein was obviously decreased compared with the model group(P<0.01).Conclusion:1.Rougantongluo Decoction has a protective effect on rat cerebral ischemia-reperfusion injury.2.Upregulation of XIAP while downregulation of Smac expression is one of the neuroprotective mechanisms.
