| Objectives: To observe the influence of dark tea on aortal inflammatory cytokines and NF-κB of hyperlipidemia rat,to discuss the mechanism of dark tea’s resistance to atherosclerosis.Methods: 60 male SD rats were randomly divided into the control group(n=10) and the model group(n=50).The control group were fed with normal diet,and the model group were fed with high fat diet for 12 weeks.Intraperitoneal injection of Vit D3 was used in the preparation of hyperlipidemia rat model.After 12 weeks, the tail blood of rats were used to detect blood lipid,so as to test if the model was successful.After the model was successful established,the rats in the model group were stratified and randomly divided,10 in each group.The control group and model group were given equal volume of distilled water.High,medium and low dose of dark tea group and Astorvastatin,respectively according to the corresponding concentration,received lavage 4 weeks, and continued high fat feed.After the experiment, using semi-automatic biochemical analyzer test in rats serum lipid levels, artery tissue pathology tests using HE staining method, using immunohistochemical method to detect the aorta inflammatory factor and NF-κB protein expression levels, inflammatory markers by q PCR method to detect the aortic and NF-κB m RNA expression level.Result: 1) By high-fat diet and intraperitoneal injection of Vit D3, can successful preparation of hyperlipidemia model, causing arterial inflammation. 2) After the intervention of the dark tea, the experimental rats serum TC, TG, LDL-c levels reduced, HDL-c levels elevated, all statistically significant(P < 0.01). After the intervention of the dark tea, the inflammatory factor and the NF-κB expression levels decreased, the difference was statistically significant(P < 0.05), different dose groups of TNF-α expression levels was statistically significant(P < 0.01), medium > low > high, ICAM-1 and IL-1β expression levels was statistically significant(P < 0.05), medium and low > high.IL-6,NF-κB P50 and P65 there are differences between different dose groups, but has not yet reached statistical significance(P > 0.05).Conclusion: 1) Dark tea can reduce serum TC, TG, LDL-c levels, increase HDL-c levels. 2) Dark tea can reduce aortic inflammatory cytokines, nuclear factor expression levels, may be it is dark tea anti-inflammatory, one of the mechanism of delay the occurrence and development of atherosclerosis. |
