Purpose: To explore the changes of rats hippocampal neurotransmitter on the transient cerebral ischemia-reperfusion injury with Buyanghuanwu Tang combined NGF,for clinical treatment of transient ischemic diseases provide more theoretical basis. Material and method:Adult male Wistar rats 280,220-280 g weight,the composite particle fed with free activities, food and water.Feeding rats 7 days later,rats were randomly divided into 7 groups according to the random number table:normal group(n=40),model control group(n=40),model group(n=40),model group sampling immediately(n=40),the treatment groups including: traditional Chinese medicine group(group A;n=40),NGF group(group B;n=40),traditional Chinese medicine+NGF group(group C;n=40).In this study,using atraumatic hemostatic clamp to clip both common carotid arteries 10 min,then recovering hemoperfusion 10 min and clipping 10 min again,so repeated this three times to establish the model of transient brain ischemia-reperfusion in rats.After operation,the treatment group A rats were given a gavage of Buyanghuanwu Tang(1.2ml/100g)·d.The treatment group B rats were intramuscular injection NGF(30?g NGF was dissolved in 2ml normal saline) once a day.The treatment group C rats were given Buyanghuanwu Tang(1.2ml/100g)·d and intramuscular injection NGF, once a day.The model group and the control group rats were given a gavage of the same amount of 0.9% normal Saline(1.2ml/100g)·d.Continually given medication for 7 days,to determine the Stroke Index score and to see the morphologic changes of neurons in hippocampus.The enzyme-histochemistry is used to study the average optical density of NOS? ACh E in hippocampus and to study the concentration of 5-HT?IL-6 in hippocampus by enzyme-linked immunosorbent assay Results:After operation,each treatment group and the model group rats appear less movement?hair disheveled?unresponsive?sleepiness and blepharoptosis and other stroke symptoms.Re- sults of light microscopy:the model group hippocampal neurons suffered significant damage. Hippocampal pyramidal cells arranged at random.Boundaries of cells are untidy.Cell volume reduced like triangular or irregular shape,and the space between the groups of cell and the surrounding tissue significantly increased.A large number of neuron loss and it is hard to find complete cell shape.There are a lot of scattered cell debris in the hippocampus.Compared with the normal group and the model control group,the number of neurons in hippocampus of the model group has significantly reduced(P<0.01).The average optical density of NOS and the concentration of IL-6 in hippocampus have significantly increased(P<0.01).The average optical density of ACh E and the concentration of 5-HT have significantly reduced(P<0.01). Compared with the model group,the number of neurons of each treatment groups were significantly increased(P<0.01).The average optical density of NOS and the concentration of IL-6 in hippocampus have significantly reduced(P<0.01).The average optical density of ACh E and the concentration of 5-HT have significantly increased(P<0.01). Conclusion:NOS?ACh E?5-HT and IL-6 are involved in the transient cerebral ischemiareperfusion injury pathological process.the treatment of each group can reduce the transient cerebral ischemia-reperfusion injury in neurons,and the effect of group C treatment significantly.
