Purpose:to study the abnormal small intestine movement is a common symptom of spleen-deficiency, but the mechanism is unclear. Vasoactive intestinal peptide (vasoactive intestinal peptide, VIP) is a kind of important ghrelin, its role in the abnormal spleen-deficiency small bowel movement. Animal groups choose only SPF 20 male SD rats, and randomly divided into healthy controls and spleen-deficiency model group (model group), each group of 10.2. Model replication and evaluation type diet dishonor and weary spleen-deficiency model, namely:within 15 d (1) feed 1 d first, fast 2 d again, all water, a total of five cycles; (2) the daily water temperature 35?37? swimming to exhaustion. Evaluation standard of weight, eat less, fatigue strength of god, the colour bent without luster.3. The small intestine movement Use charcoal propulsion method at the end of the rat small intestine propulsion rate level.4. Small intestine morphology using HE staining and electron microscope to observe the morphology of small intestine.5.Assay for VIP, cAMP and PKA ELISA was used to measure their concentration in the small intestinal tissue.6. VPAC2 Rexpression Immunohistochemical technique was used to dectect its expression in the small intestinal tissue.7.The data processing using SPSS 17.0 statistical software analysis, experimental data to adopt (X±S), using t test, P<0.05 was statistically significant. Results:1. The model evaluation Compared with those in the control group, the body mass is rudeced, food and water intakes were decreased, motion distance and vertical times were reduced, and forelimb strength were decreased, significantly, in the model group.2. The small intestine propulsion rate Compared with that in control group, the small intestine propulsion rate was significantly reduced in the model group..3. The morphological changes In the control group, the small intestinal mucosa epithelial arrangement showed that, well arranged, rich microvilli, nuclei were rods, configuration was consistent, close connection between cells were shorter, bridge structures were good.,cytoplasm in rough endoplasmic reticulum, mitochondria were rich. And that in the model group showed that, mucosal surface epithelial arranged well, cytoplasm and microvilli were rich, close connection was short.4. Changes of VIP-VPAC2R-cAMP-PKA signal transduction pathways Compared with those in the control group, the VIP level was increased, VPAC2R expression and levels of cAMP and PKA level were all decreased, significantly, in the model group.Conclusion:The abnormality in VIP-VPAC2R-cAMP-PKA pathway is involved in the reduciton of the small intestinal movement in spleen qi deficiency.
