Objective:To investigate the matrix metalloproteinase-2,9(MMP-2, MMP-9), matrix metalloproteinase inhibitor-1(TIMP-1) expression and significance in colon adenocarcinoma.Methods:Using immunohistochemical SP method,MMP-2, MMP-9 and TIMP-1 protein expression in 110 cases of colon adenoca- rcinoma and 40 cases of normal tiss--ue adjacent to adenocarcinoma were detected.Results:1?MMP-2,MMP-9and TIMP-1 protein expression rates in colon adenocarcinoma were 54.55%,59.10% and 34.55% respectively,While the positive rates in normal tissue adjacent to adenocarcinoma were 17.50%,15.00% and 42.50% respectively. The expression of MMP-2 and MMP-9 in colonic cancer tissue is higher than that in normal tissue adjacent to adenocarcinoma( ?2=16.288,22.875;P<0.05). The expression of TIMP-1 in the two groups has no obvious difference(P>0.05). 2?The expression of both MMP-2 and MMP-9 in low differentiation group were obviously higher than that in high differentiation group(P<0.05);Meanwhile,it is higher in lymph node metastasis group than that in without metastasis(P<0.05).There were significantly different(P<0.05)in the expression of TIMP-1 in groups of lymph node metastasis, the low degree of differentiation, clinical period of III-?. 3?The expression of between MMP-2 and MMP-9 was positively correlated in colon adenocarcinoma P<0.05), while The expression of TIMP-1 were negatively correlated respectively to MMP-2 and MMP-9(R=0.365, 0.297;P<0.05).Conclusion:1?The expression of MMP-2 and MMP-9 in colon adenocarcinoma tissue is significantly higher than that in normal tissue adjacent to adenocarcinoma,indicating that MMP-2 and MMP-9 were involved in the occurrence and development of colon adenocarcinoma progress. 2?In colon adenocarcinoma tissues,the high positive expression rates of MMP-2 and MMP-9,and the low positive expression rate of TIMP-1 respectively were relevant with differentiation, lymph node metastasis and clinical stage, suggesting the increase of MMP-2 and MMP-9 expression and the decrease of TIMP-1 expression with colonic cancer closely related to it's invasion and metastasis.the expressive increase of MMP-2 and MMP-9 and decrease of TIMP-1 promote colonic adenocarcinoma metastasis jointly. 3?The expression of between MMP-2 and MMP-9 was positively correlated incolon adenocarcinoma,while the expression of TIMP-1 were negatively correlated respectively to MMP-2 and MMP-9,so combined detection expression of MMP-2,MMP-9 and TIMP-1,may be used as evaluation reference index of boi- logical behavior and prognosis of colonic adenocarcinoma.
