| Objective To observe the intervention effects of Xin-Tong tai on apolipoprotein E gene knockout(ApoE-/-)mice in the process of atherosclerosis,explore the possible mechanisms and targets.Method forty-two 6-8 week old ApoE-knockout male mice were fed with high-diet for 12 weeks.Five mice were randomly sacrificed to obtain aortas for Hematoxylin and Eosin(HE)staining,and the AS model was ascertained under microscopy.then,the remaining 40 mice were randomly divided into 4 groups :model group,Xin-Tong tai Iow-dose,high-dose groups and Atorvastatin Calcium Tablets group.The C57BL/6J mice that homologous with ApoE-/-mice,were fed by food as the blank group.5groups in all.Two dose groups of Xin-Tong tai intragastric administration were designed as follow:Xin-Tong tai low dose group(3g/kg/d),Xin-Tong tai high dose group(12g/kg/d).Atorvastatin Calcium treated group(3mg/kg/d)and mice from blank and model group were given equal volume of normal saline(0.2ml/kg/d).At the end of 8 weeks,mice were sacrificed to observe serum index(TC,TG,HDL-C,LDL-C level and Lp-PLA2,ox-LDL level were measured).The aorta were separated carefully and cut into fine tissue pieces to find the pathomorphism changes by microscope.Results1.Compared with the normal group,obvious pathological changes in aorta were observed in the model group.The level of TC,TG,LDL-C,Lp-PLA2,ox-LDL were significantly increased and the HDL-C was significantly decreased(P<0.01).2.There were distinctive difference in vascular anatomy,Serum Biochemical between the model group and drug intervention groups(two dose of Xin-Tong tai groups and Atorvastatin Calcium group).plaque area and lumen area narrowing rate in the model group were larger.The blood TG,TC,LDL-C,Lp-PLA2,ox-LDL have remarkable reduced after treatment(P<0.01).3.Compared with the Xin-Tong tai low-dose group,the indexes including the lumen area,plaquearea(PA)and the level of TG,TC,LDL-C,Lp-PLA2,ox-LDL were reduced significantly in the high dose of Xin-Tong tai groups and Atorvastatin Calcium group(P<0.01).There were no significant differences among AtorvastatinCalcium group,Xin-Tong tai high dose group(P>0.05).Conclusion Xin-Tong tai can inhibit the area of atherosclerotic plaque,promote the function of arteries,So effective inhibition in dyslipidemia will decrease the level of TG,TC,LDL-C.In addition,research and trials have shown that Xin-Tong tai could prevent the pathologic progress of atherosclerosis,which could reduce content of inflammatory cytokines Lp-PLA2,and suppresses inflammation,Decreasing ox-LDL level to manage the deleterious effects of oxidative stress. |
