Purpose:To study the effects of lipid-lowering tablets on the expression of VEGF,CD31 and PI3-K in AS plaques.Methods:Forty Japanese rabbits were divided into 4 groups according to the random number table,which were blank control group,high fat model group,lipid-lowering tablets group and simvastatin group.In addition to the blank control group,the remaining groups each morning to give high fat diet 100 g,fed for 12 weeks to manufacture atherosclerosis model.After 12 weeks the lipid-lowering tablets group daily gavage 0.39g·kg-1·d-1;The simvastatin group was given daily 1.96 mg · kg-1 · d-1.The blank group and the high-fat model group were given normal saline 10 m L / day.Each group was given 4 weeks.Blood lipids were measured after intragastric administration and pathological changes were observed.The expression level of VEGF in plaques was detected by immunohistochemical staining.The expression of P13-K and CD31 was detected by immunofluorescence assay.Results:(1)Serological examination showed that the lipid-lowering tablets group compared with the high-fat model group was significantly different(P<0.05).(2)Pathological results showed that the aortic intima thickening,the surface less smooth,endothelial cell injury,medial thickening,the formation of more obvious atherosclerotic plaque;The thickness of the aortic wall was relatively uniform in the blank control group,and the intima was smooth and complete;The simvastatin group and the lipid-lowering tablets group also showed partial thickening of the endometrium,endothelial cell swelling,degeneration is not obvious,Compared with the model group,foam cells and lipid deposition were reduced to varying degrees.(3)The results of immunofluorescence showed that CD31 and PI3 K were expressed in the high fat model group,and the endometrial was thickened and the intimal surface was less smooth and incomplete.There was no positive expression of CD31 and PI3 K in the blank control group.The intima of the control group was smooth and complete without thickening.In the simvastatin group,some positive expression was seen,and the intima became thicker than the model group.In the lipid-lowering tablets group,a small amount of expression was observed,the intima was smooth and no obvious thickening.(4)The results of immunohistochemistry showed that the expression of VEGF was increased in the model group.There was no positive expression of VEGF in the normal control group.The positive expression of VEGF was observed in simvastatin group,and the expression of lipid-lowering tablets group was less.After the treatment,the levels of VEGF were significantly decreased in the simvastatin group and the lipid-lowering tablets group(P <0.01),and the difference was statistically significant(P <0.01).Compared with simvastatin group,the difference was statistically significant(P <0.05).Conclusion:(1)Lipid-lowering tablets can improve the blood lipid in rabbits,and the effect is better than simvastatin.(2)The effect of placental plaque stabilization plaque may be related to inhibition of VEGF expression in AS plaques.(3)Lipid-lowering tablets can reduce the expression of PI3 k in atherosclerotic rabbits,down-regulate the expression of plaque CD31,improve plaque composition,and stabilize plaque.
