Objective:Neuropsychiatric systemic lupus erythematosus(NPSLE)is a common cause of disability and death in patients with systemic lupus erythematosus.In this study,we used MRL/lpr mice,a kind of well-known NPSLE model,to test the effect of Jiedu Quyu Ziyin Prescription on NPSLE.Methods:Female MRL/lpr mice and the wild type C57BL/6 mice were used in our study.MRL/lpr mice were divided into three groups:(1)model control and(2)Jiedu Quyu Ziyin Prescription group.Drugs were administered continuously for 6 weeks from the 8 weeks of age.Then,experiments such as open field test,object-recognition task,forced swim test and pilocarpine acute seizure model were used to test the effect of Jiedu Quyu Ziyin Prescription on the NPSLE related symptoms.The deposition of IgG type immune complex,cortical autoantibody titers and nissl bodies were measured to evaluate the effect of Jiedu Quyu Ziyin Prescription on NPSLE related pathological changes.Results:1.There were no significant differences in total track lengths(F ?=0.1996,P=0.821)of the open field test and the recognition index of the novel object recognition task(F-0.7369,P=0.4933)among all groups.However,center-to-total track length ratio in MRL/lpr mice was larger than that in WT mice in the open field test.Jiedu Quyu Ziyin Prescription reduced the center-to-total track length ratio(P=0.033)in MRL/lpr mice.2.In the forced swim test,the floating latency of MRL/lpr mice was shorter than that of WT mice(q=7.524,DF=18,P=0.0001),and the total floating time of MRL/lpr mice was also longer than that of WT mice(q=10.13,DF=18,P<0.0001).Jiedu Quyu Ziyin Prescription prolonged the floating latency(q=4.383,DF=18,P=0.0162)and shortened the floating time(q=4.240,DF=18,P=0.0201)in MRL/lpr mice.3.Injection of pilocarpine at 200 mg·kg-1 resulted in SE in all MRL/lpr mice but none in WT mice.Jiedu Quyu Ziyin Prescription reduced the SE latency and the number of generalized seizures in MRL/lpr mice.4.The expression of intracerebral ACA(q=8.635,DF=15,P<0.0001),anti-NMDAR antibody(q=9.297,DF=15,P<0.0001)titers and IgG type immune complex(P=0.0219)in MRL/lpr mice were higher than that in C57BL/6 controls,Jiedu Quyu Ziyin Prescription reduced the level of ACA(q=4.461,DF=15,P=0.0169)and anti-NMDAR(q=3.864,DF=15,P=0.0387)antibody in MRL/lpr mice.The lgG type immune complex is similar between Jiedu Quyu Ziyin Prescription and WT group.5.Hippocampal neurons arranged loosely and their Nissl bodies were decreased in MRL/lpr mice.Jiedu Quyu Ziyin Prescription increased the number of Nissl bodies in MRL/lpr mice.In addition,the average optical density of GFAP in MRL/lpr mice was lower than that in WT mice,which may be due to the reduction of the number of astrocytes and their synapses.Jiedu Quyu Ziyin Prescription increased average optical density of GFAP in MRL/lpr mice,which may increase the number of hippocampal astrocyte and their neurites.Conclusion:Thus,our results proved direct experimental evidence that Jiedu Quyu Ziyin Prescription can relieve MRL/lpr mice NPSLE-related symptoms.Jiedu Quyu Ziyin prescription also reduced the autoantibody titers and IgG type immune complex deposition,and rescued hippocampal pathological impairment.Thus,our results suggesting Jiedu Quyu Ziyin prescription may be able to treat or prevent NPSLE.
