Objective:To investigate the effect of Alternating Current at Intermediate Frequency(ACIF)in combination with Paclitaxel on cell proliferation of MCF-7 cells in vitro.Method:Cell proliferation in culture was studied in human breast MCF-7 cell lines exposed to ACIF,Paclitaxel separately and in combinations.Survival rate of different ACIF,effects of different concentration of Paclitaxel and ACIF in combination with Paclitaxel on cell proliferation of MCF-7 cells,besides time course of recovery from 24 hour exposure to ACIF,Paclitaxel alone and in combination with ACIF were evaluated by cck8 assay;Cell apoptosis/death and cycle of MCF-7 cells were analyzed by flow cytometry.Results:1.With the increase of current(50-100mA),survival rate of human breast MCF-7 cells is more and more low,and survival rates of the experimental groups compared with control group were statistically significant differences(P<0.01).2.With the growth of concentration of Paclitaxel(0.01-1000 nmol/L),inhibition rate of Paclitaxel on MCF-7 cells becomes more and more strong,experimental groups compared with control group were statistically significant differences(P<0.01),and IC50 of Paclitaxel on MCF-7 cells is 31 noml/L.3.After MCF-7 cells exposed to ACIF(100 kHz,110mA,30 min),Survival rate of MCF-7 cells is 70±5.24%,but significantly decreased to 24±1.81%(P<0.01)in combination with Paclitaxel.4.With 24h treatment,all of the experimental groups on MCF-7 have different inhibition rates.human breast MCF-7 cells of ACIF group and Paclitaxel group compared with control group decreased by 30%.last but not least,significantly decreased by 50%(P<0.01)atattch to IC50 in combination with Paclitaxel.During the next 48h,MCF-7 cells of ACIF group compared with Paclitaxel group recovered more faster,but slower in combination with Paclitaxel.Survival rate of ACIF in combination with Paclitaxel group is minimum,only 33%(P<0.01).5.Both of ACIF and Paclitaxel group are able to promote MCF-7 cells apoptosis,but a clear increase in combination with Paclitaxel(P<0.01).6.all of the experimental groups compared with control group could induce MCF-7 cells to be arrested in S phase?G2/M phase,particularly more G2/M phase in combination with Paclitaxel group(P<0.01).conclusion:ACIF alone could inhibit MCF-7 cells proliferation and in combination with paclitaxel cancer treatment could increase chemotherapeutic efficacy.
