| Objective:Chronic obstructive pulmonary disease(COPD)is a chronic onset of complex systemic disease in airway and lung parenchyma and pulmonary vascular inflammation,chronic obviously specific clinical manifestation characteristics of airflow limitation is not fully reversible,can be progressive,and often accompanied by cardiovascular disease,diabetes,asthma and anemia and other complications.The incidence of the disease is related to environmental exposure.COPD belongs to the category of "cough","cough and asthma","lung distension","Zhi Yin" of TCM.Gubenzhike medicine,focusing on supporting healthy and strengthening Zhengqi,supplemented by Eli Quxie disease to more.This experiment have observed intervention of Gubenzhike traditional Chinese medicine on the respiratory tract and digestive tract of mice PIgR,SIgA synthesis and secretion of influence,in order to explore the effect of Gubenzhike on respiratory tract and gastrointestinal mucosal immunity;interaction between respiratory and digestive canal mucosal immune for providing a new theoretical basis to "lung regulating visceral activities".Methods:100 mice were randomly divided into 5 groups as blank control group,model control group,high dose group,middle dose group and low dose group.Except control group,other groups with cigarette smoking combined COPD model made by the method of nasal instillation of lipopolysaccharide,for a period of 60 days;then three Chinese medicine groups given of Chinese medicine with corresponding concentration by gavage,once a day for 30 days.Control group and model group were fed with drinking water.Then,we observe every group with pathological changes of mouse lung tissue morphology,PIgR and SIgA levels of mice lung BALF,intestinal mucus and lung homogenate,intestinal tissue(ELISA).Results:Pathological observation of mice model of bronchial mucosa can be seen pathology performance of COPD.The morphological changes of lung tissue were improved in different dose groups.The results of ELISA detection showed that compared with the control group(P<0.01),compared with the control group,the SIgA in bronchoalveolar lavage fluid(BALF)was significantly lower than that in the control group.The content of SIgA in the middle and low dose group of the traditional Chinese medicine was significantly higher than that of the model group(P<0.01).The middle dose group was significantly higher than the high dose group(P<0.01).Compared with the control group(P<0.01),compared with the control group,the intestinal mucus SIgA content was significantly lower than that of the control group.High,middle and low dose group and model group were significantly improved(P<0.01).The middle dose group was significantly higher than that of the high and low dose group,the difference was statistically significant(P<0.01);it is worth noting that,compared with the control group,the middle dose group of small intestinal mucus SIgA was a little higher,but the difference was not statistically significant(P>0.05).Compared with the control group,the content of PIgR in lung homogenate was significantly lower than that in the blank group(P<0.01).The content of PIgR in the middle dose group was significantly higher than that in the model group(P<0.01),while the high and low dose groups were slightly increased compared with the model group,but the difference was not statistically significant(P>0.05).The middle dose group was slightly higher than that of the high and low dose group,but the difference was not statistically significant(P>0.05);middle dose group compared with the control group,the content of PIgR is low,and the difference was not statistically significant(P>0.05).Compared with the control group,the content of PIgR in the small intestine homogenate was less than that in the blank control group,but the difference was not statistically significant(P>0.05).Compared with the model group,the middle dose group and low dose group were improved,and the difference between the middle dose group and the model group was statistically significant(P<0.01).In the small intestine of PIgR was higher than that of high dose group,low dose group and high dose group compared with significant difference(P<0.01);it is worth noting that the synthesis of PIgR content in small intestine of dose of Chinese medicine group was significantly higher than that of control group(P<0.01).Conclusion:In COPD model mice,the damage of respiratory tract mucosa and the deficiency of immune function may lead to the decrease of PIgR synthesis and SIgA secretion of respiratory mucosal immune system,intestinal mucosal immune system.It is suggesting that Gubenzhike medicine can enhance respiratory mucosa immune function of mice,and the effect of medium dose group is better than which in high and low ones.Based on the above results,it is suggested that the prevention and treatment of COPD by the traditional Chinese medicine can be realized by enhancing the mucosal immune function of the respiratory tract.The immune indexes of respiratory tract and intestinal mucosa showed high synchronization.It provides a new theoretical basis for "lung regulating visceral activities". |
