| Background: The incidence of atherosclerosis increased year by year.Recently the coronary heart disease(CHD)and acute coronary syndrome(ACS)has been concerned more and more,which is seriously harmful to life and health.With the deeper and expansion of related experiments,integrin alpha 5 beta 1 and its downstream key pathway of FAK was gradually included by researchers.Initially,studies showed that FAK has relationship with the occurrence,invasion,metastasis and prognosis of many malignant tumors,such as colon cancer,breast cancer,gastric cancer,non small cell lung cancer and so on.The biological effects of FAK are mainly dependent on the phosphoylation of key sites(Tyr397).It had been reported that FAK plays an important role in the process of angiogenesis and angiogenesis in the embryonic stage.And FAK participated in regulating migration and proliferation of endothelial cells and smooth muscle cells.Therefore,it is speculated that FAK participates in and promotes the occurrence and development of atherosclerosis.Purpose: In this study,we detected the expression of CD68,actin,CD131,FAK and phospho FAK in in experimental group(atherosclerosis group)and control group(normal artery group)by immunohistochemical,analysis the function and correlation between FAK and phospho FAK in occurrence and development of atherosclerosis.Method: 100 autopsy specimens were selected from the Department of pathology and forensic medicine of Dalian Medical University in the past 5 years.Left anterior descending artery,left coronary artery and right coronary artery were taken respectively.In this study,the main experimental methods were used in the immunohistochemical method and elastic fiber staining method,and the image analysis software was used to analyze the images.The dyeing of CD68 and actin were used to assist the localization of FAK and phospho FAK in the atherosclerotic plaque.The new blood vessels were marked by CD131,and count the density of new blood vessels detected the correlation between FAK and angiogenesis.According to the different stages of atherosclerotic plaque,the tissues were divided into two groups: the fat pattern and atherosclerotic plaque group.The coronary artery tissues were divided into A group(< 25%),B group(25%-50%),C group(50%-75%)and D group(>75%)according to the degree of stenosis.Result: 1.FAK and FAK are both expressed in cytoplasm of endothelial cell,endometrial stromal cell,macrophage,smooth muscle cell,especially in the smooth muscle cell migration to intima and calcification foci.2.The expression intensity of FAK in the smooth muscle cells decreased with increasing of the stenosis degree(P values are 0.000).The expression intensity of Phospho FAK in the smooth muscle cells and the degree of stenosis showed a tendency to decrease with the increase of the degree of stenosis.P values were: group A to group B 0.011,group A to group C 0.172,group A to group D 0.000,group B to group C 0.221,group B and group D 0.000,group C and group D 0.000.3.The number of local new born blood vessels increased with the upgrade of FAK and phospho FAK expression intensity.There is a certain positive correlation between them(R FAK=0.983,P FAK=0.000;R Phospho FAK=0.994,P phospho FAK=0.000).4.The expression of FAK and Phospho FAK in the smooth muscle cells of atherosclerotic plaque group was significantly positively correlated(R=0.904,P=0.000).In addition,FAK and Phospho FAK expression in cytoplasm of smooth muscle at the bottom of the non plaque plaque is higher than the bottom of smooth muscle,and FAK,Phospho FAK is higher than that of atherosclerotic plaque in the expression of fatty streaks endometrium near internal elastic plate interstitial.Conclusion: 1.FAK and phospho FAK participate in the occurrence and development of coronary atherosclerosis by activating the endothelial cells,promoting the migration of smooth muscle cells,and inducing the local angiogenesis in the plaque.2.FAK and phospho FAK participate in the occurrence of atherosclerosis local calcification. |
