| Significance:Diabetes is a kind of chronic metabolic disorder disease caused by inadequate insulin secretion or by the use defects.Its diagnosis mainly based on elevated blood glucose levels in the body.It is affected by genetic,immune and environmental factors.With the changes in modern lifestyles,Social pressure is getting bigger and bigger,the incidence of diabetes is increasing year by year.Not only can diabetes affect the individual’s health,the burden to the community,human health and family is also heavier.At present,Western medicine treatment of diabetes is mainly western medicine,although effect of hypoglycemic is fast,there are side effects that can’t be ignored.And acupuncture therapy can effectively interfere with type 2 diabetes(Type 2 diabetes,T2DM),as early was recorded in the "Nei Jing" and "Qian Jin Fang".In 2003,The World Health Organization also approved that acupuncture is one of the possible treatment to type 2 diabetes,access to the world’s medical community recognition."Weiwan xiushu" and "Housanli" are commonly used and experience points for the clinical treatment of T2DM,so this experiment selected these two points,to study its hypoglycemic mechanism.Objectives:In this study,JNK1-IRS1 pathway was used as the research object based on oxidative stress,one of the hotspots of diabetes mellitus.To observe the changes of serum SOD and MDA contents in serum of T2DM rats,the expression of JNK1,p-IRS1 protein and JNK1 mRNA and IRS1 mRNA in skeletal muscle,so as to evaluate the effect of electro-acupuncture on "Weiwan xiushu" and "Housanli" to T2DM rats Resistance.Finally,the possible mechanism of electro-acupuncture therapy for T2DM was analyzed from the aspects of protein and gene level.In order to further enrich the mechanism of electro-acupuncture in the treatment of type 2 diabetes.Methods:The experimental animals were 40 male SD rats.After 7 days adaptive feeding,the body mass and fasting blood glucose was measured,then random selected 8 rats as a blank control group,continue to feed the general feed.The other animals were fed with high glucose and high fat diet.2%concentration of STZ solution was injected to their abdominal cavity,according to the body mass ratio of 3 5mg/kg for modeling,and still fed with high sugar,high fat diet.All prepared model animals were randomized to measure blood glucose levels after 3 days of modeling and oral glucose tolerance(OGTT)levels after two weeks.The blood glucose>16.7mmol/L and the blood glucose>11.1mmol/L after OGTT2h were successful.Then,according to the blood glucose level the rats were divided into model group,electro-acupuncture "Weiwan xiushu" group and electro-acupuncture "Housanli" group and glimepiride group.All interventions were conducted at 18:00 pm daily,6 times a week(Monday to Saturday),for four weeks.Electro-acupuncture used continuous wave,the frequency is 5Hz,the current intensity is 3-5mA,made the experimental rats muscle slightly jitter,and didn’t occur scream as the premise,needle 20min.The body weight and fasting blood glucose were measured every 1 week.After treatments,the levels of TG,TC,HDL-C and LDL-C were measured in the serum of each group,and the changes of blood glucose,the levels of HOMA-IR and HOMA-B were calculated before and after the intervention.The levels of serum SOD and MDA in rats were measured by radioimmunoassay.The expression of JNK1 and p-IRS1 protein in rat skeletal muscle was detected by western blot method.Real-time PCR was used to detect the expression of JNK1 and IRS1 mRNA in rat skeletal muscle.Results:At the end of first week,compared with the model group,the fasting blood glucose level of the rats in the electro-acupuncture group was significantly lower than that in the model group and the difference was statistically significant(P<0.05).At the Intervention 3-4 weekend,compared with the model group,the fasting blood glucose of the rats was significantly lower than that of the model group(P<0.05).The levels of serum TC,LDL-C and TG in the rats were significantly lower than those in the model group(P<0.05).Electro-acupuncture could reduce the fasting insulin level,insulin resistance index and stomach(P<0.01).After 4 weeks of intervention,the area under the OGTT curve of "Weiwan xiushu" group and glimepiride group was significantly lower than that of the model group(P<0.05).After the intervention,the activity of SOD in the electro-acupuncture groups rats was significantly higher than that in the model group(P<0.05),and the content of MDA was significantly lower than that in the model group(P<0.05).Compared with the model group,the content of SOD and MDA in the glimepiride group was not statistically significant.The expression of JNK1 protein in the skeletal muscle of the "Wei wan xiushu" group and glimepiride group rats were significantly lower than that of the model group(P<0.05).Compared with the model group,the expression of p-IRS1 protein in the skeletal muscle of the intervention group was significantly lower than that of the model group(P<0.05),There was no significant difference between the two groups(P>0.05).There was no significant difference between the two groups(P>0.05).There was no significant difference in the p-IRS1 protein ’between the two groups(P>0.05).Compared with the model group,the expression of JNK1 gene was significantly decreased in the patients under "Weiwan xiushu"group,the posterior group and the glimepiride group(P<0.05).There was no significant difference between the intervention groups(P>0.05).Compared with the model group,the expression of IRS1 gene in each intervention group was significantly higher than that in the control group(P<0.05).There was no significant difference between the intervention groups(P>0.05).Conclusion:Under the premise of high glucose and high fat diet,electro-acupuncture of"Weiwan xiushu","Housanli" can reduce the T2DM model rats’ fasting blood glucose to a certain extent,the effect of electro-acupuncture on the "Weiwan xiushu" was significantly better than that the electro-acupuncture "Housanli" group and the glimepiride group.Electro-acupuncture has a significant effect on reducing the levels of serum TC,LDL-C and TG in type 2 diabetic rats,and the best effect was electro-acupuncture "Weiwan xiushu" point.Electro-acupuncture intervention not only reduced the fasting insulin level in T2DM model rats,but also reduced the insulin resistance index of T2DM rats to a certain extent.Among them,the effect of electro-acupuncture "Weiwan xiushu" on the improvement of HOMA-IR was the best.Electro-acupuncture "Weiwan xiushu",western medicine glimepiride has the effect of improving the glucose tolerance of type 2 diabetic rats,and then the effect of improving the impaired glucose tolerance is not obvious.Compared with the contents of SOD and MDA in the serum of each group,the electro-acupuncture "Weiwan xiushu","Housanli"can improve the oxidative stress in T2DM rats.At the level of protein and gene levels,it was shown that electro-acupuncture "Weiwan xiushu","Housanli" could reduce the expression of JNK1,p-IRS1 and JNK1 mRNA in rat skeletal muscle.The expression of IRS1 mRNA in skeletal muscle of T2DM rats was increased to a certain extent.The results were consistent with the electro-acupuncture at hypoglycemic lipid-lowering and the improvement of insulin resistance. |
