Background:Hepatitis E virus is the main pathogen of hepatitis E.According to the data from the World Health Organization in July 2015,about 20 million people were infected with hepatitis E virus all over the world every year,more than 3 million cases of acute hepatitis liver disease,and 56600 death cases related to the hepatitis E.The proportion of hepatitis E in the patients who transfused more than one time,and the organ transplantation patients were significantly higher than the general people.In recent years,transmitted hepatitis E were widely reported because of the transfusion.Objective:To uncover the hepatitis E infection status of blood donors in Nanjing,and the influence on blood safety.The genotype of HEV would be confirmed because of the antigen structure and specificity.Methods:We collected blood samples which have been tested qualified on HBV,HCV,HIV,TP and ALT.According to the current standards,and these bloods can be supplied for clinic.The HEV IgG,IgM were tested by ELISA,and if HEV IgM was positive,the HEV RNA would be tested by nucleic acid test.The ?-? different type ORF3 antigens were expressed by eukaryotic cell expression system.The ORF3 antibody was tested by ELISA which were coating the different ORF3 antigen.The genotype of HEV would be confirmed because of the antigen structure and specificity.After typing,the western blot would be carried out to confirm the antibody specificity and the accuracy.Results:5256 qualified blood samples have been tested in this study.99(1.88%)HEV-IgM positive samples and 1624(30.90%)HEV-IgG positive samples were detected by ELISA.72(1.37%)samples were both HEV-IgM and HEV-IgG positive.1(0.019%)HEV RNA positive sample was detected in all HEV-IgM positive samples.By the testing of Anti-ORF3,42(42.42%.)positive samples were detected in HEV-IgM positive samples.The genotype of HEV were genotype ? and IV.13(30.95%)samples were genotype ?,37(88.09%)samples were genotype ?,and 8(19.04%)samples were infected by both genotype ? and ?.Conclusions:HEV markers can still be detected in qualified bloods from health blood donors.The risk of hepatitis E infection cannot be excluded by current detection and standards.By the further detection,the genotype of HEV can be defined by the immune response of different ORF3 Antigen from ? to ?.The specificity of the ORF3 antibody can be confirmed through western detection,and the false positives can be identified.
