| Part Ⅰ:Effect of Albizia Chinensis(Osb.)Merr.Extract on Gastric Ulcer in RatsThe Objective was to investigate the gastric protective effect of Albizia chinensis extract on acute gastric ulcer model in rats and their effect of H~+,K~+-ATP ase of gastric parietal cells.Rats were injected intraperitoneally with indomethacin or kept under water immersion-restraint stress(WRS)to induce gastric ulcer.Test sample was given oral 30 min before induction of gastric ulcer,and gastric ulcer index and inhibitory rate were used to evaluate the effect of Albizia chinensis extract.Gastric mucosa pathological pattern in rats was observed by HE staining,the enzymatic activity of proton pump on isolated gastric parietal cells was measured,and the translocation of gastric proton pump was observed by immunofluorescence cell staining.The Albizia chinensis extract(300 mg/kg,ig)significantly inhibited gastric ulcer induced by water immersion-restraint stress or indomethacin,and the inhibitory rate was 70.0%and 75.4%.The extract inhibited the enzymatic activity of gastric proton pump,ID50 is 130μg/ml,whereas the extract had no significant effect on the translocation of gastric proton pump.Conclusion The Albizia chinensis extract(300 mg/kg,ig)have significant gastric ulcer prophylactic effect by inhibiting the enzymatic activity of gastric proton pump.Part Ⅱ:Protective Mechanism of Antidepressant Duloxetine Hydrochloride on Water Immersion Restraint Stress-Induced Gastric Ulcer in RatsThe objective was to investigate the protective effect of Duloxetine Hydrochloride in water immersion restraint stress-induced(WIRS)gastric ulceration,and to explore the antiulcer mechanism of Duloxetine Hydrochloride in the regulation of oxidative stress of brain.The contents of reduced glutathione(GSH)and MDA,as well as the activities of glutathione peroxidase(GSH-Px)and SOD in the gastric mucosa were detected after WIRS 6 h.In comparison to sham group,the content of GSH,and activity of GSH-Px and SOD in the cortex of rats have no significant change after WIRS 6 h;there was also no significant change in the content of GSH and MDA and the activity of GSH-Px in the hippocampus of rats.Duloxetine hydrochloride has significant influence neither in the content of GSH and MDA,nor in the activity of GSH-Px and SOD in rats’ cortex or hippocampus. |
