Objective: To investigate the association of the rs8052394 polymorphism of MT1A gene and the rs28366003 Polymorphism of MT2A gene With Type 2 Diabetic mellitus in kunming Han Chinese.Methods: The genotype was detected by Taqman PCR in 257 Kunming han patients with type 2 diabetes mellitus (T2DM),and 108 normal controls (NC). The genotype,allele frequency, risk factors and related clinical and biochemical data were compared among groups.Results: (1) In T2DM patients group the levels of Cr, UA, TG, FBG, HbA1C and BMI were higher than normal controls, the level of HDL-C was lower than NC group(p<0.05),the rest of the indicators there was no statistically significant difference between the two groups (p > 0.05). (2) The rs8052394 polymorphism of MT1A in comparison of T2DM and NC group: the genotype frequency of samples of each group in accordance with Hardy - Weinberg genetic law of balance (P > 0.05), in which A allele gene frequency 0.71 and G allele frequency is 0.29. A/A genotype frequency in T2DM group was significantly higher than that of NC group (x2 =20.519, p < 0.001); the A allele frequency in T2DM group was significantly higher than that of NC group (x2 - 29.213, p < 0.001).(3) The rs28366003 Polymorphism of MT2A gene in comparison of T2DM and NC group: the genotype frequency of samples of each group in accordance with Hardy - Weinberg genetic law of balance(P > 0.05), in which A allele gene frequency 0.76; G allele frequency is 0.24. G/G genotype frequency in T2DM group was higher than that of NC group (x2 = 7.167, p=0.007). There were no significant differences in A/A, A/G genotype between T2DM and NC group(x2= 4.938, p = 0.026; x2= 2.877, p = 0.090); The G allele frequency in T2DM group was significant higher than NC group (x2= 11.073, p = 0.001).(4) There were no significant differences in males and females in the rs8052394 polymorphism of MT1A gene and the rs28366003 Polymorphism of MT2A gene. (5) Logistic regression analysis showed that: the rs28366003 Polymorphism of MT2A gene had no associated with T2DM in Kunming Han Chinese, and high UA and MT1A rs8052394 A/A genotype were risk factors for occurrence of T2DM. HDL-C was protect factor for occurrence of T2DM.Conclusions: The rs28366003 Polymorphism of MT2A gene had no associated with T2DM in Kunming Han Chinese.HDL-C was protect factor for occurrence of T2DM.High UA and MT1A rs8052394 A/A genotype were risk factors for occurrence of T2DM.Objective: To explore the the association of the polymorphism of metallothionein gene with diabetic kidney disease in kunming Han Chinese.Methods: The genotype was detected by Taqman PCR in 168 Kunming han patients with type 2 diabetes mellitus (DKD0 group), 89 Kunming han patients with diabetic kidney disease (DKD group), and 108 normal controls (NC). The genotype and allele frequencies and relative clinic data were compared among groups.Results: (1) In DKD group the levels of Cr, UA, FBG and HbA1C were significant higher than NC and DKDogroup; In DKD and DKD0 group the level of TG and BMI were significant higher than NC group, and the level of HDL - C was lower than NC group. In DKD0 group the level of Cr, UA, FBG and HbA1C were significant higher than that NC group (P < 0.05). (2) The rs8052394 polymorphism of MT1A in comparison of DKD0, DKD and NC group: the genotype frequency of samples of each group in accordance with Hardy - Weinberg genetic law of balance (P > 0.05), in which A allele gene frequency 0.71 and G allele frequency is 0.29. Genotype frequency and allele frequency between each group had no statistical difference (p >0.05). (3) The rs288366003 polymorphism of MT2A in comparison of DKD0, DKD and NC group: the genotype frequency of samples of each group in accordance with Hardy - Weinberg genetic law of balance (P > 0.05), in which A allele gene frequency 0.76; G allele frequency is 0.24. G/G genotype frequency in DKD group was significant higher than that of NC and DKD0 group (?2= 17.468, p < 0.001;?2=10.286, p =0.001). G allele frequency in DKD group was significant higher than that of NC group and DKD0 group (?2= 12.637, p < 0.001).(4) Logistic regression analysis showed that: the rs8052394 Polymorphism of MT1A gene had no associated with DKD in Kunming Han Chinese , and high TG, high HbA1C and MT2A rs28366003 G/G genotype were risk factors for occurrence of DKD.Conclusions: The rs8052394 Polymorphism of MT1A gene had no associated with DKD in Kunming Han Chinese , and high TG, high HbA1C and MT2A rs28366003 G/G genotype were risk factors for occurrence of DKD.Objective: To explore the the association of the polymorphism of metallothionein gene with DM complicated with peripheral atherosclerosis in kunming Han Chinese.Methods: The genotype was detected by Taqman PCR in 87 Kunming han patients with DM complicated with peripheral atherosclerosis (AS group), and 170 Kunming han patients with type 2 diabetes mellitus (T2DM) , and 108 normal controls (NC).The genotype and allele frequencies and relative clinic data were compared among groups.Results: (1) In AS and T2DM group the level of Cr,UA, TG,FBG,BMI and HbA1C were significant higher than NC group , and the level of HDL - C was significant lower than NC group(P < 0.05). (2) The rs8052394 polymorphism of MT1A in comparison of T2DM, AS and NC group: the genotype frequency of samples of each group in accordance with Hardy - Weinberg genetic law of balance (P > 0.05), in which A allele gene frequency 0.71 and G allele frequency is 0.29. A/A genotype frequency in AS and T2DM group was significant higher than NC group (x2= 18.942,p < 0.001; x2=28.355, p =0.001). A allele frequency in AS and T2DM group was significant higher than NC group (x2=17.991,p < 0.001; x2=27.823, p < 0.001).The genotype frequencies and allele frequency between the AS group and T2DM group had no statistical difference(x 2= 4.388,p =0.111; x 2=3.876,p=0.049). (3) The rs288366003 polymorphism of MT2A in comparison of T2DM, AS and NC group:the genotype frequency of samples of each group in accordance with Hardy -Weinberg genetic law of balance (P > 0.05), in which A allele gene frequency 0.76; G allele frequency is 0.24. G/G genotype frequency in T2DM group was significant higher than NC group(x2= 16.601, p<0.001). A allele gene frequency in AS and NC groups were significant higher than T2DM group (X2= 17.266, p< 0.001, x2=9.082,p=0.003). (4) Logistic regression analysis showed that: the rs28366003 Polymorphism of MT2A gene had no associated with DM complicated with peripheral atherosclerosis in Kunming Han Chinese. HDL-C was protect factor for occurrence of DM complicated with peripheral atherosclerosis. MT1A rs8052394 A/A genotype were risk factors for occurrence of DM complicated with peripheral atherosclerosis.Conclusions: The rs28366003 Polymorphism of MT2A gene had no associated with DM complicated with peripheral atherosclerosis in Kunming Han Chinese. HDL-C was protect factor for occurrence of DM complicated with peripheral atherosclerosis.MT1A rs8052394 A/A genotype were risk factors for occurrence of DM complicated with peripheral atherosclerosis.
