| Objective:Constituents absorbed into blood and anti-influenza virus activity of medicated serum of flavonoids,phenolic acids,and alkaloids of the flowers of Trollius chinensis Bunge were evaluated,in order to provide scientific basis for further exploitation of T.chinensis.Methods:The flavonoids(37.00 mg/200 g),phenolic acids(23.05 mg/200 g),and alkaloids(20.00 mg/200 g)of the flowers of T.chinensis were administrated orally,and the drug serum were obtained at different point-in-time from inferior venca cava.LC-MS was applied to study the constitutents absorbed into blood of flavonoids,phenolic acids,and alkaloids;The MDCK cells model,infected with H1N1 influenza virus,was used for studying anti-influenza virus activity of drug serums of flavonoids,phenolic acids,and alkaloids.Result:Twelve compounds,absorbed into blood after flavonoids were administrated orally,were identified,including two original compounds(vitexin and isoswertiajaponin)and ten metabolites.Hydroxylation,the fracture of C-glycoside,reduction,ω-oxidation,the break of C ring,losing hydroxylation,sulfation,glucuronidation,glycine conjugation reactions,and so on,happened on flavonoids in vivo;The seven compounds,after oral administration of phenolic acids,were identified,including one original compounds(veratric acid)and six metabolites.There were main N-oxidation,demethylation,hydroxylation,ω-oxidation,sulfation,glycine conjugation reactions on phenolic acids in vivo;Only five metabolites,absorbed into blood after alkaloids were administrated orally,were identified.demethylation,ω-oxidation,losing hydroxylation,sulfation,glycine conjugation reactions happened on alkaloids in vivo.The drug serums of flavonoids,phenolic acids,alkaloids,and extracts possessed anti-influenza virus activity,and the point-in-time of the strongest antiviral activities of them were 90,15,45,and 25 min,respectively.Conclusion:Constituents absorbed into blood were the metabolites mainly,and original compounds were vitexin,isoswertiajaponin,and veratric acid,after flavonoids,phenolic acids,and alkaloids were administrated orally.Previous study showed most the metabolic reactions of phenolic acids and alkaloids happened at about 25 min,after extracts of the flowers of T.chinensis were administrated orally,and the anti-influenza virus activity was the strongest at 25 min,illustrating that the antiviral effective components were from phenolic acids and alkaloids mainly.In this study,the point-in-time of the strongest antiviral activities of phenolic acids and alkaloids were approximate with that of extracts of the flowers of T.chinensis,and most metabolites of phenolic acids and alkaloids were detected at the point-in-time.Therefore,the effective components of the flowers of T.chinensis are the metabolites and original compounds of phenolic acids and alkaloids in this experiment. |
