| The construction of simulated human network model,which integrated the existing knowledge of human living,is of great importance for promoting the understanding the rules of human living and motivating the emerged knowledge of human complex system.In the post genome era,the biological data presented an explosive growth in quantity,the unstructured or semi-structured data on one hand provided the data basis for human modelling and "omic" research,on the other hand,the high complexity and multidimensional property of the biological data made it so hard for the utilization of big data.in allusion to this situation,researchers in life science have attempted to utilize the biological data for model construction and made inspiring progress so far.However much of the work is partial model constructed with only one type structured data or just the visualization of the data.The poor model construction efficiency and little information contained in the models indicate that the method of big data and fragment information utilization and modelling with integrated information remain a unsolved question.Aiming at the integration and comprehensive utilization of scattered biological and big data,this paper presented a method to process the unstructured proteomic data,metabolomic data,reactomic data into a standard form and link the data with reactions under the guidance of Entity Grammar System.The different data first integrated to establish the human reaction map,then the cell specific protein expression data was integrated to construct the cell type specific network model,the cell type specific networks are linked by the messengers and receptors to establish a human whole body model.The human whole body modelconsist of 1596 pairs of reacptors and messengers,15718 proteins and 5811 reactions and 83 cells.Based on the exist knowledge of life science,this paper discussed how the different cell type specific network interact with each other.Thus proved the cell specific network represented the composition and function of the cell.Furthermore,the interactions between tissues and organs are also discussed.The theory that human whole body model is a multi-level model which contains modules is verified.Additionally,this paper also explored the drug mechanism analyzing and drug reposition based on the good simulation of human cell reactions.This paper explored the potential pharmacological effect of anti-hypertension and anti-diabetes of topiramate and the new indication of chlorpromazine.The good simulation result proves the practical value of the whole body model.The human whole body model integrated data from proteomics,metabolomics and reactomics in different form with the help of Entity Grammar System.This paper also provided a data integration method by constructing a whole body model。The model itself also has practical value and can be used in drug development,drug mechanism analysis and drug reposion and other biological related fields as a reference or a tool. |
