| Objective:To observe the effect of electroacupuncture on protein and gene expression of peritoneal macrophages in atherosclerotic rabbits,and to explore the mechanism of acupuncture on atherosclerosis and provide experimental evidence for acupuncture treatment of atherosclerosis.Methods:New Zealand rabbits,male,26,were fed at the SPF laboratory animal center.After one week of adaptive feeding,the rats were randomly divided into blank group(n = 7)and model group(n = 19).The model group was fed with high fat diet,and the corresponding blank group was fed with ordinary feed without special intervention.After 4 weeks of high fat feeding,the model group was subjected to balloon injury of carotid artery,followed by high fat feeding for 4 weeks.One of the blank group and the model group was used to observe the pathological changes of carotid atherosclerotic plaques by HE staining to determine whether the model was successful.Eighth weeks into a mold,the rats were randomly divided into model group,acupuncture group and Western medicine group,6 rats in each group,Neiguan electroacupuncture group(bilateral),Zusanli(bilateral),Guan Yuanxue EA,20min/ time,1 times a day,western medicine group were treated with Atorvastatin Calcium Tablets(1mg/kg/d)20ml gavage daily.1 times,were treated for 4 weeks.At the end of the treatment,the rabbits were anesthetized and the blood samples were collected from the ear vein.To observe the morphologic changes of rabbit carotid atherosclerotic plaque pathology HE staining;the expression of hand was detected in rabbit blood lipid level;the expression level of Western blot was used to detect the expression of macrophage CD36 protein;to detect the expression of RT-PCR receptor expression of macrophage CD36 m RNA.Results:1 HE stainingCompared with the blank group,the model group was significantly impaired arterial intima,intima of atherosclerotic plaque was observed,inflammatory cell infiltration and a large number of foam cells,rough electro acupuncture group and Western medicine group of vascular wall,a small amount of foam cells and a little plaque;compared with the model group,electroacupuncture group and Western medicine group were significantly reduced,showed that the effective electro acupuncture group and Western medicine group;compared with the western medicine group,were observed under light microscope there were not obvious difference.2 the influence of electro acupuncture on the expression of TC,TG,LDL and HDL in the blood of rabbits with atherosclerosis.Compared with the control group,the expression of the model group,electroacupuncture group and Western medicine group,TG lipids TC and LDL significantly increased,the expression of HDL was decreased,the difference was statistically significant(P<0.01);compared with the model group,electroacupuncture group and Western medicine group the expression of TC,TG and blood LDL were decreased,HDL expression was significantly increased,with statistical significance the difference(P<0.01);compared with the western medicine group,acupuncture group TC,TG LDL,blood lipid and the expression of HDL was not statistically significant(P>0.05).3 electroacupuncture on the expression of CD36 m RNA in peritoneal macrophages in atherosclerotic rabbits.Compared with the control group,the expression level of model group,electroacupuncture group and Western medicine group CD36 m RNA receptor of peritoneal macrophages was significantly increased,the difference was statistically significant(P<0.01);compared with the model group,electroacupuncture group and Western medicine group the expression of receptor of peritoneal macrophage CD36 m RNA levels were significantly lower,the difference was statistically significant(P<0.01);compared with the western medicine group,the expression of electro acupuncture group of peritoneal macrophage receptor CD36 m RNA level was not statistically significant(P>0.05).4 electroacupuncture on the expression of CD36 protein in peritoneal macrophages of atherosclerotic rabbits.Compared with the control group,the expression level of model group,electroacupuncture group and Western medicine group receptor macrophage CD36 protein increased significantly,the difference was statistically significant(P<0.01);compared with the model group,electroacupuncture group and Western medicine group the expression of receptor of peritoneal macrophages CD36 protein levels were significantly lower,the difference was statistically significant(P<0.01);compared with the western medicine group.The expression of EA group peritoneal macrophage receptor CD36 protein level was not statistically significant(P>0.05).Conclusion:1 Electro acupuncture can reduce atherosclerotic plaque of carotid artery in atherosclerotic rabbits.2 Electro acupuncture can reduce the expression of CD36 in peritoneal macrophages of atherosclerotic rabbits,which may be one of the mechanisms of acupuncture regulating atherosclerosis3 Electroacupuncture can improve the blood lipid level of atherosclerosis. |
