Based On KKS And ENOS/NO Signaling Pathways, Electroacupuncture Was Used To Protect Renal Function In Prehypertensive Rats

Purpose: Under the guidance of the preventive treatment of diseases of traditional Chinese medicine theory,Acupuncture and moxibustion in the early prevention and treatment of hypertension development,effectively blocking hypertensive target organ damage and complications associated with a definite curative effect.This experiment with high salt diet induced Dahl salt sensitive for prehypertension model rats,electroacupuncture Zuzanli points,Quchi points observing the influence of electricity for rats blood pressure,kidney function,kidney shape,application of PCR array technology to detect DS immune and inflammatory related gene expression,q PCR,WB and Elisa method to detect KKS,e NOS/NO related signal pathway expression in rats,aims to find the curative prehypertension kidney protection mechanism,provides the experimental basis for the clinical acupuncture and moxibustion prevention health care.Material and method: Male salt resistance only as blank group 10 rats,salt sensitive rats 30 according to random number table method is divided into model group,acupuncture group,the acupuncture points.Rats after unified measuring blood pressure feed conventional adaptive feed for a week,then use containing 8% Na Cl high-salt feed,daily monitoring of blood pressure in rats.Rats blood pressure to prehypertension standards(120-139/80-89 mm Hg),according with the standard model,then stoping high-salt feed and starting cupping treatment.Normal group and model group only a day to be fixed,not do acupuncture treatment;Electric acupuncture group take Zuzanli points,Quchi points on both sides of filiform needle acupuncture,depth of about 5 mm,SDZ-type II electronic needle therapy apparatus,output wires at both ends respectively connected with the lateral Zuzanli points,Quchi points stimulation parameters: the density wave,sparse wave 1 Hz,dense wave 5 Hz;Thin wave time for 10 seconds,dense wave time to 15 seconds,adjust current rat tail or lower extremity twitch slightly brown.Once per day,treatment for 20 minutes at a time.Six consecutive days for a period of treatment and rest 1 day,a total of 5 weeks treatment;Non acupoint group in the site selection of meridian courses after less reins,take 10-15 mm onbilateral iliac crest,next to the midline open after a fixed 20 mm segment control point,treatment with electric acupuncture group.After the treatment,take blood anesthesia for abdominal aorta of rats,collect serum urea nitrogen,creatinine,uric acid content in rats.Take the left kidney tissue,PCR array detection related immune and inflammatory gene expression,q PCR,WB and Elisa to detect expression of related signal pathways;HE dyeing observation DS in the rat kidney tissue pathological morphology.Results: 1.Electric acupuncture can regulate prehypertension specificity kidney partial immune and inflammatory gene expression.PCR array experiment results show that in 84 inflammatory autoimmune related genes in model group expression,gene expression significantly raised a total of 10 after cupping treatment,including Bcl6,Ccl19,Ccl4,Csf1,Cxcl10,Cxcl9,Cxcr4,Il1 rn,Tirap,Tollip;Expression model group,significantly lower after treatment with acupuncture expression of genes,a total of 2,including Il1r1,Kng1,these genes mainly related to cell apoptosis,expression of inflammation,etc.2.Electric acupuncture can affect hypertension early kidney specificity KKS related signaling pathway.The experimental results show that the electric acupuncture to specific influence KKS signaling pathways,reduce prehypertension DS rats kidney Kng m RNA expression,higher Klk and B2 R m RNA expression.Thus affect the content of NO synthetase,further induce the NO,play a role of prehypertension kidney protection.3.Electric acupuncture can adjust prehypertension DS renal function in rats.The experimental results show that the electric acupuncture can significantly reduce the prehypertension DS rats UREA and CREA,UA content,protect prehypertension kidney function.4.Electric acupuncture can specificity to adjust the rat kidney Ca M/e NOS/NO signaling pathways.The experimental results show that the electric acupuncture Zuzanli points,Quchi points can be higher specificity,e NOS gene expression in the rat kidney Ca M,increased e NOS protein content and increase the NO concentration,so as to achieve expansion blood vessel,protection of prehypertension kidney function.5.Electric acupuncture can regulate rat kidney NO specificity/c GMP/PKG signalingpathways.The experimental results show that the electric acupuncture can higher specificity s GC,PKG gene expression in rats,early diastolic hypertension rats renal blood vessels,give play to the role of hypertension early kidney protection.Conclusion: 1.Electric acupuncture can protect the prehypertension DS renal function in rats.Research shows that: the electric acupuncture Zuzanli points,Quchi points can reduce the content of serum urea nitrogen,creatinine,uric acid,protect Dahl salt sensitive rat kidney function,prevent pre-hypertension target organ damage.2.Cupping by adjusting the KKS and e NOS/NO blood pressure signal pathway,thereby promoting vasodilation,play a role of kidney protection.Research shows that: the electric acupuncture Quchi,Zuzanli to activate kidney KKS signal pathway,so as to change the intracellular calcium ion concentration,activate the Ca M guide e NOS/NO signaling pathways,thus affecting the c GMP/PKG content,in the end diastolic renal small vessel.This may be a cupping to protect one of prehypertension renal function normal operation mechanism.
