Liver cancer which ranks as the second malignant tumor in China is one of the ten common diseases in the world.The current study shows that the growth and proliferation is closely related to the level of the tumor cell apoptosis.Therefore,in the treatment of the cancer,it is an important strategy to induce the occurrence of the tumor cell apoptosis.So,at the same time,it is key to find a kind of effective anticancer substance in the nature to prevent the occurrence of the liver cancer.Tea is one of the three beverages in the world,and our country is a big tea producer with a long history of planting tea and a wide tea-planting region.From ancient time to today,tea is popular in china.Tea includes the ingredients which are active,antioxidant,and anti-inflammatory,antiviral and so on.At the same time,tea can prevent the proliferation of the HepG2;the anti-cancer effect of tea is more and more concerned.Because of the different processing technology of tea,the origin is different and variety are different,especially the processing technology is closely related to its composition change,such as green tea,black tea and Fuzhuan tea.Typical tea species represent the different processes of non-fermentation,semi-fermentation and total fermentation,but the substances in different teaseffect on the activeness of the malignant tumor and the function of the tumor cells have not been reported.Therefore,the thesis focuses on green tea,half-fermented tea,black tea and Fuzhuan tea.By exploring the impact on the activeness of the tumor cells and the function of the active substances,do the research to record the level of the tumor cell apoptosis.In the study,we take the way of experiment outside the body and choose the exponential phase HepG2 cells in good condition as the subject.The substances ed from the four teas are used as the test materials.MTT is taken to test the materials' impact on the HepG2,with the help of florescence microscope and FCM to observe and measure the cell morphology,period the apoptosis rate of the cells,active oxygen and the change of membrane potential,at the same time measure the protein expression with the help of the western blotting.Following are the key research findings.(1)The four categories of green tea,half fermented tea,black tea and Fuzhuan tea of polyphenols content of the results show that the total phenol content in tea:green tea>half fermented tea>Fuzhuan tea>black tea,high performance liquid chromatography(HPLC)analysis of green tea and half fermented tea was the most abundant Epigallocatechin gallate(EGCG),while the content of Caffeine(CAF)was the highest in Fuzhuan tea and black tea.(2)Green tea,half fermented tea,black tea and Fuzhuan tea obviously restrain the growth of HepG2 and the dose is closely related to the cell growth.And could some obvious cell apoptosis features such as the chromatin Pyknosis,the serious cell deformation,and so on.Various rough extract can stop the cell period and promote the' cell apoptosis,effects of the four teas restrain the proliferation of HepG2:green tea>half fermented tea>Fuzhuan tea>black tea.(3)After studying the function of green tea seducing and promoting the apoptosis of HepG2.The level of the active oxygen inside HepG2 obviously increases,mitochondrial membrane potential obviously declines,the expression of protein Cyt-c promoting the cell apoptosis rises up,Caspase-3/9 become active,the expression ratio of Bax promoting the cell apoptosis and Bcl-2 restraining the cell apoptosis obviously gets up,the expression of p53 gets up,the expression of PARP inside the cell declines.(4)The four tea prevent the liver cell growth by blocking the cell period to increase the rate of cell apoptosis among which the green tea could inhibit the growth of HepG2 cells by blocking the cell cycle and inducing the mitochondrial apoptotic pathway in a dose-dependent manner.
