Photodynamic therapy,as an important treatment for skin cancer and subcutaneous tumor tissue,plays an important role in cancer therapy.The main principle of photodynamic therapy is the use of photosensitizer,in the light to produce a large number of ROS,and the use of its toxins to kill cancer cells,to achieve the purpose of curing the tumor.At present,in the process of tumor photodynamic therapy,a high concentration of intracellular GSH,which reduces the effects of photodynamic therapy.And the lack of oxygen of tumor area also affects the levels of ROS,which effects of photodynamic therapy.In response to these problems,we developed two types of materials?Cu-MOF,Mn-MOF?.Cu-MOF significantly reduced the level of GSH in the cell,enhanced the ROS level,and enhanced the photodynamic therapy.In addition,the O2content in the tumor area was increased by Mn-MOF,the ROS level was improved,and the photodynamic therapy was enhanced.The main work of this paper includes the following:1.To reduce the effect of high concentration of GSH on photodynamic therapy in cells,we developed nano-MOF with copper as the active center to enhance the photodynamic therapy by direct adsorption of GSH in cells,increase of ROS in cells.We use simple way of synthesis,was synthesized based on the structure of Cu-porphyrin?Cu-MOF?,and through ROS probe and GSH kit of Cu-MOF generate ROS and adsorption ability of GSH was determined,then Cu-MOF uptaken by breast cancer cells,by MTT test and flow cytometry test examines the Cu-MOF through adsorption of GSH induced apoptosis,compared the results solved with CPT and BSO,searched the Cu-MOF under the condition of light,produce ROS,coordinated the ability of inducing cell apoptosis,finally the Cu-MOF are applied to the treatment of breast cancer tumors in mice.Results showed that the material can be individually apoptosis induced by adsorption of GSH,at the same time can generate ROS under illumination,the synergy,enhance the effect of PDT,aggravate cell apoptosis and inhibit tumor growth.The work provides new ideas for other tumor therapy methods.2.In view of the tumor area lack oxygen environment also affects the photosensitizer produce reactive oxygen species level,will affect the photodynamic therapy.On the basis of previous work,we use simple way of synthesis,was synthesized based on the structure of porphyrin?Mn-MOF?measure the amount of oxygen was produced by Mn-MOF,and the amounts of generated 1O2 and ROS were tested with the Mn-MOF after irradiation with a 650 nm laser,by MTT test examines the Mn-MOF induced apoptosis,finally the Mn-MOF are applied to the treatment of breast cancer tumors in mice.Results showed that the material can produce O2,at the same time can generate ROS under illumination,effectively converted O2 to ROS,aggravate cell apoptosis and inhibit tumor growth,can effectively solve the problem of solid tumors parts of a lack of oxygen,have important significance on the treatment of tumor.
