The Pharmacological Study And Clinical Efficacy Of Shuanghua Yuyang Decoction In The Treatment Of Wet Heat And Poisonous Behcet's Disease

Objective:Discussion on mechanism of Shuang hua yu yang fang(SHYYF)in treatment Behcet's disease based on Network Pharmacology,and observe the clinical efficacy and safety of the treatment of Behcet's disease with damp-heat toxin accumulation syndrome by the SHYYF.Methods:1.The Methods of research on network pharmacologyThe ingredients of the drugs were obtained at TCM@Taiwan,and use the PubChem Website to transform the ingredients to SMILES format,if any SMILES cannot be got,the Chemdraw software could be used.Then predict the targets of the drugs at Superpred Server through SMILES format.Through the TTD database to obtain the targets of BD,The two compared to get the active ingredients and targets.To construct a network of the active ingredients and targets by Cytoscape sofeware.The end,explore the KEGG pathway and Genetic function of targets by ClueGO sofeware.2.The Methods of research on the clinical efficacy38 Patients coming from Department of rheumatology of Xiyuan hospital CACMs in March 2017 to March 2018.The people were treated by the SHYYF for 8 weeks.Before and after treatment,observe the symptoms and signs score changes,record the laboratory indicators(ESR,CRP),BDCAF2006 and safety indicators and adverse reactions,and analyse the results finally.Results:1.The Results of research on network pharmacology(1)There are 48 active ingredients of the SHYYF can act on 17 targets of BD,Jin-yinhua and Danggui can act the most targets,so it may be the important drugs in this decoction,similar to the Jun medicine.Lugen,Zisuye,Zhebeimu act the less targets,simila rto the Minister medicine.Shengyimi,Xuanshen,Chishao,Daqingye,Shenggancao act the le a-st targets,similar to the Adjuvant medicine.(2)Jinyinhua has 4 active ingredients can act CCR5,indirect regulate the balance of Th1 and Th2,inhibit the development of inflammation reaction.Xuanshen can act TLR4,curb the generation of TNF-a to slow down the disease.(3)KEGG pathway showed that the decoction can act NF-KB pathway,restrain the inflammatory reaction.(4)The Genetic functions showed the SHYYF can reduce synthesis of nitric oxide,relieve the vascular inflammatory reaction and the injury of vascular endothelial cell.2.The Results of research on the clinical efficacyThe total efficiency of the group is 81.82%.The symptoms and signs scores'comparisones within group pre and post treatment shows that the differences in oral ulcers,joint pain,skin lesions,irritability,fatigue,bitter taste,dry-mouth,deep-colored urine,dry stool,thin stool,insomnia and dreaminess,total integral,ESR,CRP,BDCAF2006 have obvious statistical significance.In terms of security,the Chinese-western medicine may cause leucocyte count raise and hepatic injury,and the only Chinese herbal medicine has higher safety.Conclusion:the results of the network pharmacology showed that the SHYYF plays a role through targets,KEGG pathway and the Genetic functions to treatment BD.The results of the clinical observations showed the SHYYF can improve clinical symptoms and reduce inflammation,and has curative effcet with fewer adverse reactions.
