ObjectiveThis study intends to establish an animal model of blood-heat psoriasis and to explore the therapeutic effects and mechanisms of Liangxuexiaoban Decoction on it.In this study,male BALB/c mice were used as subjects,and a mixture of dry ginger ginger and imiquimod cream was used for modeling blood heat psoriasis,and various factors and modeling conditions were investigated.The influence of the duration and stability of the blood-heat psoriasis animal model was initially summarized to optimize the overall behavioral changes and syndrome characteristics of the blood-heat psoriasis animal model.On this basis,different doses of Liangxue Xiaoban Decoction were intervened to observe its intervention on the skin lesions of blood-heat psoriasis animal model in BABL/c mice and its influence on the immune system.Methods(1)125 SPF grade male BABL/c mices,body weight 20-25g,10-12 weeks of age,pre-experimental feeding 3-5 days,free drinking water diet.They were divided into blank group,model group,high blood heat dose group,blood heat medium dose group,blood heat low dose group,25 in each group.Vaseline was smeared on the back of the blank group,and the model group and the blood heat low,medium and high dose groups were small.Ratsroutinely cast imiquimod cream.The low-does,medium-does,and high-dose blood-supply groups were given different doses of ginger decoction.The doses were 2.25g/kg,4.5g/kg,and 9g/kg,respectively,starting from the first day of the experiment and ending at the 14th day.The psoriasis model was observed from day 1 to day 28 of the experiment.(2)112 male BABL/c mices weighing 20-25 g,10-12 weeks of age,were fed 3 days before the experiment and were given a free drinking water diet.Then divided into blank group,model group,Liangxuexiaoban decoction low dose group,middle dose group,high dose group,compound Qingdai capsule group,Tripterygium glycosides group a total of 7 groups,16 in each group.According to the experimental process,the first model re-administration treatment.The blood heat model lasted for 14 days.On the 11th day,the imiquimod cream was applied until the end of the experiment on the 22nd day.The material was taken on the 19th and the 23rd day;the treatment was performed from the 15th day to the 22nd day.The blank group and model group were given physiological saline,compound Qingshao capsule group 2g/kg/d administration intervention,Tripterygium glycoside group 10mg/kg/d.The low,medium and high doses of Liangxue Xiaoban Decoction were 27.5g/kg,55g/kg,110g/kg,once a day,0.2ml each time.(3)The overall performance of each group of mice was observed:the effect of drug intervention was evaluated by weight,PASI score,epidermal thickness,HE staining,enzyme-linked immunosorbent assay,and flow cytometry.Results(1)The best model for the blood-heat psoriasis animal model is dry ginger,cinnamon decoction gavage 14 days,began to apply imiquimod cream on the 7th day,15 days combined After the completion of the mold,psoriatic skin lesions were at peak stage,and the skin lesions gradually decreased with time.The skin lesions of the model group and the low-and high-blood-heat group were similar,and the skin lesions of the high-dose blood group were the highest.Severe,the best dose is 9g/kg;(2)The method of applying imiquimod cream on the back and intragastric administration of dried ginger cinnamon decoction can prolong the skin lesions and increase the degree of skin lesions,which is an effective form of blood-heat psoriasis.Modal method.The skin lesions of the mice were observed with the naked eye,and red spots,white scales,and skin were thickened to varying degrees on the skin;in the HE-stained pathological sections,the mice in the model group and the blood-heat low-,medium-,and high-dose groups had epidermal growth.Thick,dermal inflammatory cell infiltration and capillary hyperplasia;PASI score,the most serious skin lesions in the high-dose blood group,related indicators of thyroid T3,T4 increased,spleen index increased.(3)The dynamic observation of each index found that the model of blood-heat psoriasis in BABL/c mice has obvious characteristics.Animals usually develop within 2-3 days,and the lesions gradually become serious.The PASI scores in each group begin to rise,reaching the peak of the disease within about 7 days,and then enter the chronic protracted period,the skin lesions have subsided,and the pathological lesions are maintained at a certain level.s level.(4)Compared with Tripterygium Glycosides Group and Compound Qingqi Capsules,the high dose group of Liangxuexiaoban Decoction has a more obvious protective effect on the skin lesions of BABL/c mice.The different doses of Liangxue Xiaoban Decoction had different inhibitory effects on the animal models of blood heat psoriasis,and the comprehensive regulation effect of the high dose group was better than that of the middle and low dose groups.Liangxuexiaoban Decoction can regulate the balance of Thl/Th2 and Th 17/Treg lymphocytes by reducing TNF-? and IL-17.Conclusions(1)The animal model of blood heat psoriasis was established through the improvement of model building of blood-heat psoriasis model in BABL/c mice.This model of blood heat syndrome can simulate psoriasis from the aspect of disease characteristics.The development of the disease,prolonged onset,more serious skin lesions.(2)Liangxuexiaoban Decoction has a good therapeutic effect on BABL/c mouse model of blood-heat psoriasis,can inhibit the skin injury in the acute phase of animal models,and has a certain adjustment effect on the immune system.Heat and cooling blood,reduce the body's metabolism of thyroid hormones.
