Insomnia is a sleep disorder characterized by frequent and persistent difficulty in sleeping or difficulty in maintaining sleep,which is characterized by insufficient sleep satisfaction.In traditional Chinese medicine insomnia is called "Bu Mei".The nomenclature of insomnia in different periods is different,and the angle of treatment is different too.Therefore,this study intends to organize and study the ancient and modern literature.to analysis and induction insomnia from the name,TCM etiology and pathogenesis,treatment and syndrome and other aspects.On this basis,combined with the progress of modern medical research is helpful to discusse the inhibitory protective effect of GABA energy conduction system on insomnia.Besides,the Liver depression and qi stagnation is regarded as the core pathogenesis of insomnia.The treatment method is take "Sini-suanzaoren",which palys a role on dispersing stagnated liver qi for relieving qi stagnation and nourishing blood and softening the liver.Then evaluate the effects of drugs on sleep efficiency of insomnia rats from a macro perspective.And the expression level of receptor proteins and genes related to GABA can be detected by molecular biology technology.Finally,we will make clear the action target of"Sini-suanzaoren" in the treatment of insomnia and provide research ideas for clinical treatment.The theoretical researchStudy 1 The knowledge of insomnia in traditional Chinese medicineThis study analyzed and studied the name,pathogenesis,and treatment of insomnia to make clear the different names in in different periods.By this researche we know the pathogenesis of insomnia is of one sort or another,like being invaded by exogenous pathogen,emotional disorder,eating and drinking without temperance,aged,overfatigue,weak after illness and so on.The pathogenesis of insomnia can be summarized as the imbalance of yin and Yang,disharmony of Ying Wei Qi and blood,Dysfunctional State of the Viscera,the phlegm.On the rule of treatment,according to the situation of the times and the characteristics of drug use by different doctors have different methods.Study 2 Study on the correlation and treatment of insomnia with liver qi stagnation syndromeThis study discusses relationship between insomnia and liver which has physiological characteristics like controlling dispersion and storing the blood.Due to the characteristics of the times,results show emotional factors have become the main cause of insomnia.And the occurrence of insomnia will further affect the liver function.Eventually all these factors lead to liver qi stagnation syndrome become the core pathogenesis of insomnia.Based on the main physiological characteristics of the liver and the closely relationship between the heart and the liver,so that the main treatment method is soothing the liver.At the same time the heart should be given consideration to avoid deficiency of blood that can induce worse insomnia.The representative Chinese herbal medicine to treat insomnia is "Sini-suanzaoren".The experimental studyStudy 1 Drug intervention study of Insomnia model ratsObjective Rebuilt and evaluate insomnia model rat.Methods Rats were injected intraperitoneally with PC PA to rebuilt insomnia model,and to observe general state,spontaneous activity,the difference of sleep latency and total sleep duration by the open field test,the elevated plus maze and pentobarbital sodium.The rats of PCPA insomnia model were randomly divided into five groups:The model group,the medicine group,Sini group,Suanzaoren group,Sini-suanzaoren group,also set up a blank control group.Each group was given the drug for 7 days,and the model group and blank control group were given normal saline lavage 7 days in a row.The rats general activity state and weight increment were observed,and the difference of sleep latency and total sleep duration was observed by using sodium salt in the rats.Results The control group rats are in a good mental state,sensitive,hair luster,weight increased significantly.The model group rats with a dry and dull hair were irritability and angular,and the water quantity of rats increased obviously.Compared with the model group,the mental and activity state of the medicine group and the traditional Chinese medicine were better.Compared with the control,the weight of the model group was very significantly decreased(P<0.01),Compared with the model group,the weight of the medicine group,Suanzaoren group,Sini-suanzaoren group was very significantly increased(P<0.01).Compared with the control,the difference of sleep latency of the model group was very significantly increased(P<0.01).Compared with the model group,the difference of sleep latency of the medicine group,Sini group,Suanzaoren group,Sini-suanzaoren group was significantly or very significantly decreased(P<0.05 or P<0.01)Compared with the control,the total sleep duration of the model was significantly decreased(P<0.05).Compared with the model,the total sleep duration of the medicine group,Sini group,Suanzaoren group,Sini-suanzaoren group was significantly or very significantly increased(P<0.05 or P<0.01).Conclusions Rats injected intraperitoneally with PCPA can successfully rebuilt insomnia model rat,and the insomnia rats were unhappy,less static dynamic and spontaneous activity.Dispersing stagnated liver qi for relieving qi stagnation and nourishing blood and softening the liver can decrease insomnia rats the difference of sleep latency and increase the total sleep duration.Study 2 Effect of "Sini-Suanzaoren" on hypothalamus cells morphology and GABA conduction system related target protein of Rats with InsomniaObjective To study the hypothalamus cell morphological changes of insomnia rats,and the mechanism of GABA conduction system related target protein after treated with"Sini-Suanzaoren".Methods Observe the hypothalamus cells morphological of the rats by H.E.stain method.Detection the content of GABA?GABAARal?GABAAR?2 and NKCC1 of rats hypothalamus with Immunohistochemical method and protein imprinting method.Results Compared with the control,the hypothalamus cells morphological of the model shown some degree of variation,and the other group shown the improved significantly.The Immunohistochemical test revealed that,compared with the control,the content of GABA?GABAAR?1?GABAAR?2 of model was significantly decreased(P<0.05),compared with the model,the content of the medicine group,Sini group,Suanzaoren group,Sini-suanzaoren group was significantly or very significantly increased(P<0.05 or P<0.01).Compared with the control,the content of NKCC1 of model was significantly increased(P<0.05),compared with the model,the content of the medicine group,Suanzaoren group,Sini-suanzaoren group was significantly decreased(P<0.05).The protein imprinting method test revealed that,compared with the control,the content of GABAAR?1?GABAAR?2 of model was significantly decreased(P<0.05),compared with the model,the content of the medicine group,Sini-suanzaoren group was significantly or very significantly increased(P<0.05 or P<0.01).Compared with the control,the content of NKCC1 of model was very significantly increased(P<0.01),compared with the model,the content of the medicine group,Sini group,Suanzaoren group,Sini-suanzaoren group was very significantly decreased(P<0.01).Conclusions The H.E.stain method revealed that the hypothalamus cells morphological of the model shown some degree of variation,and the other group shown the improved significantly.The results improve that the medicine,sini scattering,suanzaoren prescriptions and sini-suanzaoren prescriptions can improve the hypothalamus cell mutation status.The Immunohistochemical test and the protein imprinting method test revealed that,the content of GABA?GABAAR?1 GABAAR?2 of hypothalamus was decreased,and NKCC1 was increased,sini-suanzaoren can increase the expression of protein of GABA?GABAAR?1?GABAAR?2,and decrease the NKCC1.The process can produce inhibitory nerve protective effect and improve the state of insomnia rats through strengthening the combination of GABA and its receptor.Study 3 Effect of“Sini-Suanzaoren”on GABA conduction system related target protein mRNA of Rats hypothalamus cells with InsomniaObjective To study GABA conduction system related target protein mRNA of rats hypothalamus cells with Insomnia after treated with "Sini-Suanzaoren".Methods Detection the transcription of GABAAR?1 mRNA?GABAAR?2 mRNA and NKCC1mRNA of rats hypothalamus with RT-PCR technique.Results The RT-PCR technique revealed that,compared with the control,the transcription of GABAAR?1 mRNA of model was significantly decreased(P<0.05),compared with the model,the transcription of the medicine group,Suanzaoren group,Sini-suanzaoren group was significantly increased(P<0.05).The GABAARy2 mRNA and NKCC1mRNA transcription of each group rats were no significant difference,but the model GABAARy2 mRNA was obviously decreased,and other groups were obviously increased.The model NKCC1mRNA was obviously increased,and other groups were obviously decreased.Conclusions Sini-suanzaoren prescriptions can intervention of insomnia,this process is closely related to the up-regulation of the mRNA express of GABAAR?1 and GABAAR?2,but reduce the mRNA express of NKCC1 which play a role of inhibitory neural protection.
