Objective: To investigate the effects of Panax quinquefolium Ye Zaogan(PQS)on the expression of peroxisome proliferator activated receptor gamma(PPAR)and inflammatory factors in rats with focal cerebral ischemia reperfusion injury(PPAR),Neuroprotective effects of PQS from the perspective of anti inflammation.Method: Healthy male SD rats were randomly divided into 5 groups,namely sham operation group,model group,PQS high-dose treatment group 200mg/kg group,PQS medium dose treatment group 100mg/kg and PQS low-dose treatment group,group 50mg/kg.The rat model of focal cerebral ischemia reperfusion injury was prepared by modified Zea Longa middle cerebral artery occlusion(MCAO)method.After ischemia,2h was pulled out to achieve reperfusion.Objective To observe the effect of PQS on the behavioral changes induced by 24 h after cerebral ischemia reperfusion in rats with MCAO,Detection of myeloperoxidase(MPO)activity in brain tissue of ischemic rats by spectrophotometer,Enzyme linked immunosorbent assay(ELISA)for the detection of tumor necrosis factor-? alpha in ischemic brain tissue(TNF-?)and interleukin-6(IL-6)content,Western blot was used to detect the expression of PPAR? and nuclear factor kappa B(NF-?B)protein in the ischemic brain tissue of rats.Result: PQS can significantly improve the neurological symptoms and reduce the water content of cerebral ischemic tissue after 24 h reperfusion in rats with focal cerebral ischemia(2h);PQS significantly promoted the expression of PPAR in brain tissue after focal cerebral ischemia and reperfusion in rats with 2H,and PQS significantly inhibited the expression of inflammatory factors in the brain tissue of rats with focal cerebral ischemia after 2H reperfusion 24 h.Conclusion:PQS has protective effect on focal cerebral ischemia reperfusion injury in rats,the mechanism may be related to the effect of PQS on the expression of PPAR? in brain tissue,which can inhibit the expression of inflammatory factors and reduce inflammatory response.
