Root hair is tubular structure and formed by the root epidermal cells through selectively differentiation and polarized growth development.The occurrence of root hairs can effectively increase the surface area of the root system,at same times,root hair is thought to increase plants in water and nutrients absorption and utilization.So,root hair cell development and root hair polarized growth is crucial for building integral Root system architecture(RSA).Some recent studies have shown that the growth of root hairs is a very typical polar growth process.The actin cytoskeleton participates in the polarity growth process of root hair by Physiological activity of providing a molecular track for vesicle trafficking and cytoplasmic streaming.ACTIN DEPOLYMERIZING FACTOR(ADF)participate in root hair development and growth by modulating the rate of actin filament turnover.However,the molecular mechanisms about actin cytoskeleton involved in root hair growth is still not clear,need further study.So,we do basic Research on function of AtADF8 and AtADFll in Arabidopsis thaliana by the methods such as Real-time PCR,fluorescent Location and protein purification.In order to reveal the molecular mechanisms about ADF8/11 in the regulation process of root hair development and growth,and has achieved major results are as follows:1.RT-PCR and RT-QPCR results showed that AtADF8 have highly expressed in roots and almost no expression in Arabidopsis other tissues.ADF11 have expressed in different organ and highest expression in root tissue.2.We applied Prokaryotic expression vector successful Purifying ADF8 and ADF 11 proteins with HIS tags.These proteins can be used in high or low-speed co-sedimentation experiments in vitro,and as the foundation for exploring specific functions ADF8/11 protein.3.Confocal fluorescence micrographs showing Sub-cellular localization of ADF8-eGFP and ADF11-eGFP,We almost certain ADF8-eGFP and ADF11-GFP co-localizes with actin filaments in root hair cells and root hairs.4.We successfully construct ADF8/11 RNAi interference vector and obtain its Arabidopsis transgenic lines,providing the basis material for the study of gene function.In summary,AtADF8/11 have highly expressed in roots and co-localized with actin filaments in root hair cells.The mutants of AtADF8/11 will be a good material in subsequent experiments and will be the foundation for research of ADF8/11 gene function.Our study not only reveal the molecular mechanisms in the regulation process of root hair development,but also contributes to providing a theoretical basis and scientific foundation for the development of root and the actin cytoskeleton.
